为建立HPLC测定不同产地龙血竭中3种黄酮类成分的方法,选择了广西、云南产9个批次的龙血竭,测定了其中龙血素A、龙血素B、剑叶龙血素B的含量.采用Thermo BDS Hypersil C18色谱柱(4.6 mm×250 mm,5μm),流动相乙腈-1%冰醋酸,梯度洗脱,流速1.0 m L·min~(-1),柱温40℃,检测波长278 nm.3个药效成分在45min内达到基线分离,线性关系良好.9个批次龙血竭中龙血素A、龙血素B、剑叶龙血素B的平均含量分别为:0.47%,0.96%,0.44%;平均加样回收率分别为:99.6%,103.2%,100.5%;RSD分别为:1.20%,0.65%,1.30%.该实验方法操作简便,结果准确,重复性和稳定性良好,可为龙血竭的质量控制提供参考.  相似文献

建立反相高效液相色谱法测定龙血竭中剑叶龙血素C含量的方法,以Phenomenex C18反相键合硅胶色谱柱为固定相,水-乙腈(33:67)为流动相,流速1ml/min,检测波长为211nm;柱温35℃;用外标法定量。结果表明,剑叶龙血素C在42.85~342.8μg进样范围内有良好线性关系(r=0.9996),方法回收率为97.87%。本方法简便、准确,重复性好,可作为龙血竭中剑叶龙血素C的含量测定方法。  相似文献

研究灌胃给予大鼠龙血竭后,P糖蛋白抑制剂维拉帕米对其有效成分龙血素A、B、C在大鼠血浆中的药物动力学影响.将SD大鼠随机分为对照组和抑制剂组并单次给药,对照组灌胃给予大鼠5 g/kg龙血竭,抑制剂组联合给予大鼠维拉帕米(1 mg/kg)和龙血竭(5 g/kg).收集两组相同系列时间的血浆样本,采用HPLC-MS/MS的方法对龙血素A、B、C在大鼠血浆中的浓度测进行检测,求算两组血浆样本药物动力学参数.与对照组相比,抑制剂组大鼠龙血素A、B、C的血药浓度-时间曲线下面积分别增加109.4%,78.5%,22.8%,血药峰浓度分别增加69.6%,115.0%,42.1%,龙血素A、B的达峰时间均延长、龙血素C的达峰时间无变化,三者的生物半衰期(T0.5)均变小,说明P糖蛋白抑制剂能够引起龙血素A、B、C在大鼠体内的血浆药物动力学参数变化,龙血素A、B、C均有可能为P糖蛋白的潜在底物.   相似文献

从中药龙血竭中提取、分离、纯化制备龙血素B,经UV、IR、NM R、M S和TLC、HPLC等方法测定结构和纯度。结果得到结构正确、纯度达到99%以上、符合中药化学对照品技术要求、可作为龙血素B的化学对照品。  相似文献

为建立龙血竭指纹图谱质量控制方法,采用HPLC法测定,液相色谱仪Agilent 1100,色谱柱为Waters Xterra C184.6×250 mm,5μm;流动相为0.1%的磷酸溶液和乙腈梯度洗脱,流速为0.7 mL/min,检测波长为280 nm,理论板数按参照物(龙血素B)峰计算不低于6000.实验结果表明,10批成品指纹图谱与对照指纹图谱计算相似度,其结果均大于0.90.该方法稳定可行,可用于龙血竭质量控制.  相似文献

 对人工诱导龙血竭与野生龙血竭的化学成分进行比较研究,为人工诱导龙血竭是否能替代其野生龙血竭提供依据,解决龙血竭资源极其紧张的问题.采用HPLC法.选用Waters Nova-Pak C18色谱柱(φ3.9mm×150mm,5μm),检测波278nm,柱温40℃,进样量10μL,以乙腈-0.4%磷酸(体积比30:70)为流动相;流速1.0mL/min.结果: 龙血竭和人工诱导龙血竭均具有相对保留时间14.2,19.9min的2个共有峰,且HPLC峰形相似度较高.  相似文献

研究传统名贵傣药龙血竭中有效成分龙血素B在大鼠体内重要组织的分布情况,同时探讨长期模拟失重效应对龙血素B分布的潜在影响.将SD大鼠分为正常重力组和模拟失重效应组,采用21 d大鼠尾悬吊方法模拟长期失重效应,单次灌胃给予大鼠25 mg/kg龙血素B,于给药1 h后收集心、肝、脾、肺、肾、胃、肠、脑、睾丸和骨骼肌10种组织,采用HPLC-MS/MS方法测定各组织中龙血素B质量分数.在正常重力组中,龙血素B在肝、胃分布较多,其次是肾、肠、心、脾、骨骼肌和肺,脑中最少.与正常重力组相比,模拟失重效应组大鼠肝中龙血素B的质量分数显著升高47.7%(p<0.05),脑中质量分数上升5.4倍(p<0.05);在肠和肾中质量分数显著下降52.7%和22.0%(p<0.05);在大鼠胃、心、肺、脾、睾丸和骨骼肌中质量分数无显著变化.龙血素B的分布具有明显组织特异性,长期模拟失重效应显著改变龙血素B在大鼠肝、肠、脑、肾等重要组织的分布.   相似文献

利用毛细管区带电泳技术(CZE)研究5个龙血竭有效单体与牛凝血酶(B-Thr)、29碱基凝血酶核酸适配体(Apt29)之间的相互作用.电泳条件为有效长度是40 cm的非涂层石英毛细管,气压进样3.45 kPa,进样时间5 s,分离电压+15 kV,分离温度15℃,检测波长分别为214 nm(牛凝血酶体系)和330 nm(核酸适配体体系).结果表明,龙血素A和B-Thr之间的结合常数Kb为4.73×104 L/mol,白藜芦醇、龙血素B、7,4'-二羟基黄酮、龙血素C与B-Thr没有显著结合;龙血素B和7,4'-二羟基黄酮与核酸适配体的结合常数Kb分别为1.98×104,1.83×104 L/mol,白藜芦醇、龙血素A、龙血素C与核酸适配体之间没有表现出明显的结合现象.本文结果对进一步研究龙血竭在凝血酶上的潜在结合位点,以及核酸适配体能否向凝血酶靶向转运龙血竭,提供了科学数据支持.   相似文献

The discovery of the prolific Ordovician Red River reservoirs in 1995 in southeastern Saskatchewan was the catalyst for extensive exploration activity which resulted in the discovery of more than 15 new Red River pools. The best yields of Red River production to date have been from dolomite reservoirs. Understanding the processes of dolomitization is, therefore, crucial for the prediction of the connectivity, spatial distribution and heterogeneity of dolomite reservoirs.The Red River reservoirs in the Midale area consist of 3~4 thin dolomitized zones, with a total thickness of about 20 m, which occur at the top of the Yeoman Formation. Two types of replacement dolomite were recognized in the Red River reservoir: dolomitized burrow infills and dolomitized host matrix. The spatial distribution of dolomite suggests that burrowing organisms played an important role in facilitating the fluid flow in the backfilled sediments. This resulted in penecontemporaneous dolomitization of burrow infills by normal seawater. The dolomite in the host matrix is interpreted as having occurred at shallow burial by evaporitic seawater during precipitation of Lake Almar anhydrite that immediately overlies the Yeoman Formation. However, the low δ18O values of dolomited burrow infills (-5.9‰~ -7.8‰, PDB) and matrix dolomites (-6.6‰~ -8.1‰, avg. -7.4‰ PDB) compared to the estimated values for the late Ordovician marine dolomite could be attributed to modification and alteration of dolomite at higher temperatures during deeper burial, which could also be responsible for its 87Sr/86Sr ratios (0.7084~0.7088) that are higher than suggested for the late Ordovician seawaters (0.7078~0.7080). The trace amounts of saddle dolomite cement in the Red River carbonates are probably related to "cannibalization" of earlier replacement dolomite during the chemical compaction.  相似文献

There are numerous geometric objects stored in the spatial databases. An importance function in a spatial database is that users can browse the geometric objects as a map efficiently. Thus the spatial database should display the geometric objects users concern about swiftly onto the display window. This process includes two operations:retrieve data from database and then draw them onto screen. Accordingly, to improve the efficiency, we should try to reduce time of both retrieving object and displaying them. The former can be achieved with the aid of spatial index such as R-tree, the latter require to simplify the objects. Simplification means that objects are shown with sufficient but not with unnecessary detail which depend on the scale of browse. So the major problem is how to retrieve data at different detail level efficiently. This paper introduces the implementation of a multi-scale index in the spatial database SISP (Spatial Information Shared Platform) which is generalized from R-tree. The difference between the generalization and the R-tree lies on two facets: One is that every node and geometric object in the generalization is assigned with a importance value which denote the importance of them, and every vertex in the objects are assigned with a importance value,too. The importance value can be use to decide which data should be retrieve from disk in a query. The other difference is that geometric objects in the generalization are divided into one or more sub-blocks, and vertexes are total ordered by their importance value. With the help of the generalized R-tree, one can easily retrieve data at different detail levels.Some experiments are performed on real-life data to evaluate the performance of solutions that separately use normal spatial index and multi-scale spatial index. The results show that the solution using multi-scale index in SISP is satisfying.  相似文献

本文叙述了对海南岛及其毗邻大陆边缘白垩纪到第四纪地层岩石进行古地磁研究的全部工作过程。通过分析岩石中剩余磁矢量的磁偏角及磁倾角的变化,提出海南岛白垩纪以来经历的构造演化模式如下:早期伴随顺时针旋转而向南迁移,后期伴随逆时针转动并向北运移。联系该地区及邻区的地质、地球物理资料,对海南岛上述的构造地体运动提出以下认识:北部湾内早期有一拉张作用,主要是该作用使湾内地壳显著伸长减薄,形成北部湾盆地。从而导致了海南岛的早期构造运动,而海南岛后期的构造运动则主要是受南海海底扩张的影响。海南地体运动规律的阐明对于了解北部湾油气盆地的形成演化有重要的理论和实际意义。  相似文献

The explosive growth of the Internet and database applications has driven database to be more scalable and available, and able to support on-line scaling without interrupting service. To support more client's queries without downtime and degrading the response time, more nodes have to be scaled up while the database is running. This paper presents the overview of scalable and available database that satisfies the above characteristics. And we propose a novel on-line scaling method. Our method improves the existing on-line scaling method for fast response time and higher throughputs. Our proposed method reduces unnecessary network use, i.e. , we decrease the number of data copy by reusing the backup data. Also, our on-line scaling operation can be processed parallel by selecting adequate nodes as new node. Our performance study shows that our method results in significant reduction in data copy time.  相似文献

R-Tree is a good structure for spatial searching. But in this indexing structure,either the sequence of nodes in the same level or sequence of traveling these nodes when queries are made is random. Since the possibility that the object appears in different MBR which have the same parents node is different, if we make the subnode who has the most possibility be traveled first, the time cost will be decreased in most of the cases. In some case, the possibility of a point belong to a rectangle will shows direct proportion with the size of the rectangle. But this conclusion is based on an assumption that the objects are symmetrically distributing in the area and this assumption is not always coming into existence. Now we found a more direct parameter to scale the possibility and made a little change on the structure of R-tree, to increase the possibility of founding the satisfying answer in the front sub trees. The geographic information service is enabled by the advancements in general Web service technology and the focused efforts of the OGC in defining XML-based Web GIS service. Based on these models, this paper addresses the issue of services chaining,the process of combining or pipelining results from several interoperable GIS Web Services to create a customized solution. This paper presents a mediated chaining architecture in which a specific service takes responsibility for performing the process that describes a service chain. We designed the Spatial Information Process Language (SIPL) for dynamic modeling and describing the service chain, also a prototype of the Spatial Information Process Execution Engine (SIPEE) is implemented for executing processes written in SIPL. Discussion of measures to improve the functionality and performance of such system will be included.  相似文献
