An endo-β-1,4-glucanase (EG) was purified from the hindgut of an Australian mound-building termite, Coptotermes lacteus. The hindgut extract had a peak separate from those for extracts obtained from the salivary glands and the midgut based on sephacryl S-200 gel chromatography, and also demonstrated an origin different from the endogenous EGs of the termite itself. The recovery was further purified by SDS-PAGE, and its N-terminal amino acid sequence analyzed. This showed high homology to EGs from glycoside hydrolase family (GHF) 7. PCR-based cloning methods were applied to the hindgut contents of C. lacteus and individual protozoan symbionts from C. formosanus. cDNAs encoding putative EGs homologous to GHF7 members were then identified. The functionality of one of the putative proteins was confirmed by its expression in Escherichia coli. Summary The elimination of (14C)-DMN after i.p. injection intoXenopus was measured, as was the metabolism in vitro of (14C)-DMN by liver fromXenopus and 9 other amphibian species. In view of its rapid elimination from the body and low rate of metabolism byXenopus liver in vitro, DMN is unlikely to be toxic or carcinogenic inXenopus.This work was supported by a grant from the Cancer Research Campaign. A part of the gene coding for a halophilic serine protease from a halophilic archaeumHaloferax mediterranei R4 was amplified by PCR and its 672 nucleotide sequence was determined. Tentative translation to the amino acid sequence suggested that the enzyme was quite similar to halolysin produced by another halophilic archaeum strain 172P1. Nucleotide sequences of 16S rRNA encoding genes from 9 halophilic archaea were determined. Alignment of 19 sequences known so far showed that there are more than 20 positions carrying bases or deletions specific for each halobacterial genus:Halobacterium, Haloarcula, Haloferax, andHalococcus.  相似文献

Lipopeptaibols are members of a novel group of naturally occurring, short peptides with antimicrobial activity, characterized by a lipophilic acyl chain at the N-terminus, a high content of the turn/helix forming α-aminoisobutyric acid and a 1,2-amino alcohol at the C-terminus. The amino acid sequences range from 6 to 10 residues and the fatty acyl moieties from 8 to 15 carbon atoms. The peptide portion of lipopeptaibols can be shorter than those of the nonlipidated peptaibols that range from 10 to 19 amino acid residues. The longest peptides fold into a mixed 310/α helix, whereas the shortest peptides tend to adopt a β-turn/sheet structure. Using solution methodologies, a series of analogues of trichogin GA IV was synthesized which allowed determination of the minimal lipid chain and peptide main-chain lengths for the onset of membrane activity and exploitation of a number of spectroscopic techniques aimed at determining its preferred conformation under a variety of conditions and investigating in detail its mode of interaction with, and its effect on, the phospholipid membranes. Rossbank functioned from 1840 to 1854 as one of a chain of British Colonial Observatories which combined with European and Asian observatories in the study of terrestrial magnetism. It was established in Hobart, Tasmania, by the Governor of Van Diemen's Land, Sir John Franklin, and Captain James Clark Ross, R.N., commanding H.M. ships Erebus and Terror. The history and operation of the Rossbank Observatory is related, its instruments described, and the results discussed.

Biographical notes on the Observatory staff, with lists of its archives and instruments are provided. A number of previously unpublished pictures show the Observatory in use and the surviving buildings and instruments today.  相似文献

A bitter tasting cyanoglucoside, sarmentosin, was isolated from an aposematic Apollo butterfly,Parnassius phoebus, and from its plant-host,Sedum stenopetalum. The content of sarmentosin in the body tissues was as high as 500 g/insect, suggesting a defensive role for this substance; a high concentration was detected in the wings. Summary A macromolecular fraction from the culture fluid ofAchromobacter metalcaligenes showing the activity of the Haemophilus-X factor was investigated for its content of porphyrin derivatives. Porphyrin methylesters were directly prepared from the lyophilized material and then separated by thin layer chromatography. Protoporphyrin and iron protoporphyrin could be isolated in amounts which correlated with the growth-promoting activity forHaemophilus influenzae.  相似文献

Summary The antimitotic activity of nimbidin, a drug from the plantMelia azadirecta indica, was assessed by its effect on the meristematic cells of onion root tips. The effect was almost similar to those of colchicine and vinca alkaloids. Recovery trials showed that the drug induces lethal damage in a considerable proportion of treated cells and may hence have applications in cancer chemotherapy.  相似文献

It is well known that the co-chaperone p23 regulates Hsp90 chaperone activity in protein folding. In Plasmodium falciparum, a putative p23 (Pfp23) has been identified through genome analysis, but its authenticity has remained unconfirmed since co-immunoprecipitation experiments failed to show its interaction with P. falciparum Hsp90 (PfHsp90). Thus, recombinant Pfp23 and PfHsp90 proteins purified from expressed clones were used in this study. It was clear that Pfp23 exhibited chaperone activity by virtue of its ability to suppress citrate synthase aggregation at 45°C. Pfp23 was also shown to interact with PfHsp90 and to suppress its ATPase activity. Analyses of modeled Pfp23-PfHsp90 protein complex and site-directed mutagenesis further revealed strategically placed amino acid residues, K91, H93, W94 and K96, in Pfp23 to be crucial for binding PfHsp90. Collectively, this study has provided experimental evidence for the inherent chaperone function of Pfp23 and its interaction with PfHsp90, a sequel widely required for client protein activation.  相似文献

Summary A toxin causing paralysis was detected in the starfishAstropecten latespinosus. The toxin was purified by a method consisting of charcoal treatment and chromatography on CM-Sephadex C-25 and Bio-Rex 70. The toxin was identified as tetrodotoxin by its behavior in thin-layer chromatography and electrophoresis and its1H-NMR spectrum.Acknowledgment. In its infancy the British Association for the Advancement of Science derived a good deal of its inspiration from the writings of Francis Bacon. But the pursuit of Baconian policies brought with it attendant dangers which critics from Charles Dickens to the Times were not slow to magnify. Although the situation was further complicated by the sensitiveness of institutional Christianity at the start of Victoria's reign, some of the hazards which the Association endured had to be accepted simply as consequences of the attempt to provide a secure platform for British science.  相似文献

Summary The quinone primin (1) and its quinol miconidin (2) which occur naturally in Miconia spp. (Melastomataceae), were synthesized and then tested as potential antifeedants against 6 insect species. Summary Hagenius brevistylus lost mass by evaporation in a moderately desiccating environment at the rate of 20.4 mg h–1, and died of desiccation in less than 1 day at a body mass of 79.8% of their normally hydrated mass. It was estimated thatHagenius minimally would have to consume the equivalent of 60% of its body mass each day to meet its daily water requirements. Summary Ascorbic acid mediated a small but significant degradation of methylmercury to inorganic mercury in goldfish (Carassius auratus) and reduced the toxicity of mercuric chloride despite its substantial conversion into organic form. Acetol and methylglyoxal are intermediates of the intrahepatic metabolism of acetone leading to pyruvate formation. In hepatocytes prepared from fasted streptozotocin-induced diabetic mice, net glucose production could be measured from methylglyoxal but not from acetone or acetol. Insulin increased glucose formation from methylglyoxal in a concentration-dependent manner, whereas it was ineffective when pyruvate was used as substrate. Drug oxidation, as evidenced byp-aminophenol formation from aniline, was enhanced by methylglyoxal, and insulin proved to be stimulatory in this case as well. It is concluded that insulin might be involved in the regulation of glucose formation from methylglyoxal, but its mode of action is not yet clear.  相似文献

Bark beetles,Ips typographus andPityogenes chalcographus, attracted by synthetic or natural pheromone to Norway spruce logs,Picea abies, preferred to colonize uninfested logs rather than logs occupied by these beetles, probably as a means of avoiding intra-and interspecific competition. The aggregation pheromone components ofP. chalcographus, chalcogran and methyl (E, Z)-2,4-decadienoate, inhibited the attraction response ofI. typographus to its pheromone components (methyl butenol andcis-verbenol), while the converse was not true. Adriaan van Roomen published an outline of what he called a Mathesis Universalis in 1597. This earned him a well-deserved place in the history of early modern ideas about a universal mathematics which was intended to encompass both geometry and arithmetic and to provide general rules valid for operations involving numbers, geometrical magnitudes, and all other quantities amenable to measurement and calculation. ‘Mathesis Universalis’ (MU) became the most common (though not the only) term for mathematical theories developed with that aim. At some time around 1600 van Roomen composed a new version of his MU, considerably different from the earlier one. This second version was never effectively published and it has not been discussed in detail in the secondary literature before. The text has, however, survived and the two versions are presented and compared in the present article. Sections 1–6 are about the first version of van Roomen’s MU the occasion of its publication (a controversy about Archimedes’ treatise on the circle, Sect. 2), its conceptual context (Sect. 3), its structure (with an overview of its definitions, axioms, and theorems) and its dependence on Clavius’ use of numbers in dealing with both rational and irrational ratios (Sect. 4), the geometrical interpretation of arithmetical operations multiplication and division (Sect. 5), and an analysis of its content in modern terms. In his second version of a MU van Roomen took algebra into account, inspired by Viète’s early treatises; he planned to publish it as part of a new edition of Al-Khwarizmi’s treatise on algebra (Sect. 7). Section 8 describes the conceptual background and the difficulties involved in the merging of algebra and geometry; Sect. 9 summarizes and analyzes the definitions, axioms and theorems of the second version, noting the differences with the first version and tracing the influence of Viète. Section 10 deals with the influence of van Roomen on later discussions of MU, and briefly sketches Descartes’ ideas about MU as expressed in the latter’s Regulae.  相似文献
