Two tetrahydropyridine alkaloids, arecaidine and guvacine that are characteristic of betel nut (Areca catechu L.) have been detected in the deliberately stained labial surface of female teeth excavation on Rota, Mariana Islands. This was accomplished using selected ion monitoring techniques in conjunction with gas chromatography/electron impact-mass spectrometry. These alkaloids were not present on the buccal surface of the teeth and indicate the use of betel nut to effect the staining.  相似文献

It has been suggested that a common sex pheromone composition may account for interspecific sexual interactions observed with certain moths in the Arctiidae. In this study, it is demonstrated that the sex pheromones released by females of the Scarlet Tiger Moth,Callimorpha dominula L., and the Cinnabar Moth,Tyria jacobaeae L., have similar activities and elute at the same retention time on analysis by coupled gas chromatography (GC)-electrophysiology with males from each species. Peak enhancement on GC, chiral GC and coupled GC-mass spectrometry using authentic compounds show that the sex pheromone for bothC. dominula andT. jacobaeae is (3Z,6Z,9S,10R)-9,10-epoxyheneicosa-3,6-diene.  相似文献

Summary Mitochondrial DNA (mtDNA), isolated from worker honeybee larvae, was digested by each of seven 6-base restriction enzymes. Only one enzyme (BglII) showed a mtDNA difference between the three tested races (Apis mellifera carcia, A. m. ligustica, A.m. caucasica). BothA.m. carnica andA.m. ligustica showed the same pattern, differing fromA.m. caucasica. The degree of fragment pattern similarity revealed that there is only a small level of mtDNA variation between the three races tested. This is in line with previous investigations of enzyme polymorphisms.  相似文献

Summary The metabolism of the trichothecene 4-monoacetoxyscirpenol by intact gut tissue was determined in the fungus-feeding Nitidulid,Carpophilus hemipterus (L.) and the non-fungus-feeding caterpillars, the fall armyworm,Spodoptera frugiperda (J. E. Smith) and the corn earworm,Heliothis zea (Boddie). The primary metabolite was the hydrolysis product scirpentriol. The amount of metabolism by theC. hemipterus larvae was ca 10 times that of the caterpillars on a per mg protein basis, suggesting metabolic adaptation for feeding on fungi that may contain mycotoxins.Acknowledgment. Summary In living antennae of the silkworm mothBombyx mori L. the pheromone compound (E, Z)-10,12-hexadecadienol and hexadecanol are enzymatically converted to their corresponding fatty aldehydes, acids and long-chain fatty acid esters. The pheromone is completely degraded at high rates in the antennae of freshly hatched moths. The polar volatile [3H]metabolites exclusively consist of tritiated water. The half-life of the pheromone is about 2.5 min in males and 0.5 min in females. Summary Influenza A viruses grown in different animal or human cells retain their antigenic make-up as tested by the usual immunological assays. With the aid of aSambucus nigra (L.) extract containing its lectins the viruses can be distinguished after one single passage in a different cell type by a change in their hemagglutinating properties. Binding of such lectins to influenza viruses may be a means for a more subtle classification, relating to the host cell origin of the virus.  相似文献

Summary Treatment of neonatal mice with an antiviral factor, (AVF), obtained from the leaves ofMelia azederach L. protected them against lethal encephalitis caused by Tacaribe virus inoculation. The degree of protection obtained varied from 66% to 100% depending on the virus dose. Similarly, administration of AVF to nursing mothers protected their offspring from developing virus encephalitis. AVF does not directly inactivate Tacaribe virus; it inhibits an early step (s) in the replication process in cell cultures.Dedicated to Prof. Luis Leloir on his 80th birthday.Acknowledgment. Leptothorax (Nesomyrmex) spininodis andL. (N.) echinatinodis workers lay pheromone trails when returning from food sources to their nests. Workers from within the nest begin following the trail almost immediately upon the successful forager's return and do not require a leader. The trail remains effective for about four minutes under our laboratory conditions but it is unclear whether the trail alone is sufficient to induce recruitment or whether additional signals, perhaps emitted within the nest, are involved. Pheromone trails of this type have not previously been documented for leptothoracine ants.  相似文献

A comparative analysis of 40 Trypanosoma cruzi L1Tc elements showed that the 2A self-cleaving sequence described in viruses is present in them. Of these elements, 72% maintain the canonical 2A motif (DxExNPGP). A high percentage has a conserved point mutation within the motif that has not been previously described. In vitro and in vivo expression of reporter polyproteins showed that the L1Tc2A sequence is functional. Mutations within certain L1Tc2A sequences affect the efficiency of the cleavage. The data indicate that the L1Tc2A sequence may be influencing the L1Tc enzymatic machinery determining the composition and level of the translated products. The residues located immediately upstream of the 2A consensus sequence increase the cleaving efficiency and appear to stabilize the relative amount of translated products. Legionella (L.) pneumophila, the causative agent of Legionnaires disease, is an intracellular pathogen of alveolar macrophages that resides in a compartment displaying features of endoplasmatic reticulum (ER). In this study, we show that intracellular multiplication of L. pneumophila results in a remarkable decrease in MHC class I expression by the infected monocytes. During intracellular multiplication, L. pneumophila absorbs ER-resident chaperons such as calnexin and BiP, molecules that are required for the correct formation of the MHC class I complex. Due to reduced MHC class I expression, stimulation of allogeneic blood mononuclear cells was severely inhibited by infected host cells but cytotoxicity of autologous natural killer cells against Legionella-infected monocytes was not enhanced. Thus, reduced expression of MHC class I in infected monocytes may resemble a new immune escape mechanism induced by L. pneumophila.Received 22 November 2004; received after revision 27 December 2004; accepted 5 January 2005  相似文献

Several novel type of lipopeptides were synthesized and evaluated for their ability to stimulate non-specific resistance againstLeishmania donovani infection. Peritoneal macrophages isolated from young male hamsters treated with muramyl dipeptide (MDP) and various synthetic lipopeptides (6 mg/kg i.p.) 7 days earlier, were cultured in vitro and challenged 24 h later withL. donovani promastigotes. One lipopeptide, Central Drug Research Institute (CDRI) compound 86/450, exhibited significantly higher immunostimulatory activity than MDP. Its prophylactic activity was further confirmed in hamsters by giving 2 split doses of 3 mg/kg of the compound spaced at 2 weeks, i.e. on day –7 and +7 of challenge withL. donovani amastigotes. The prophylactic effect lasted for 7 days following the last treatment with compound 86/450. The antileishmanial action of sodium stibogluconate (SAG) was also found to be enhanced by 16% in hamsters primed with compound 86/450.CDRI Communication No. 5034.  相似文献

Summary A methanolic extract ofCitrus unshiu induces oviposition by females of aCitrus-feeding swallowtail butterfly,Papilio xuthus L. The chemical factors responsible for stimulating oviposition were isolated and characterized as 5-hydroxy-N-methyltryptamine, adenosine, vicenin-2, narirutin, hesperidin and rutin. An artificial blend of these six components elicited significant oviposition behavior, apparently identical to that induced by contact with intactCitrus leaves.  相似文献

Summary LD50 values as well as time-dependent parameters of granulosis virus infections were determined at different times during the last larval instar (L5) of the codling moth,Cydia pomonella L., induced either for pupation or for diapause. Current models state that insect peripheral oscillators are directly responsive to light, while mammalian peripheral clock genes are coordinated by a master clock in the brain via intermediate factors, possibly hormonal. We show that the expression levels of two circadian clock genes, period (per) and Par Domain Protein 1 (Pdp1) in the peripheral tissue of an insect model species, the linden bug Pyrrhocoris apterus, are inversely affected by contrasting photoperiods. The effect of photoperiod on per and Pdp1 mRNA levels was found to be mediated by the corpus allatum, an endocrine gland producing juvenile hormone. Our results provide the first experimental evidence for the effect of an endocrine gland on circadian clock gene expression in insects. In 6-day-old females ofBlattella germanica, the activity of corpora allata (CA) was inhibited in vitro by juvenile hormone III (JH III). Effective doses (281.5 and 375.4 M in the medium) were somewhat higher than (although of the same order of magnitude as) the estimated intraglandular concentration of JH III at this age, and they induced about 45% inhibition of hormonal release and a significant intraglandular accumulation of JH III and methyl farnesoate. The results suggest that autoinhibitory mechanisms operate in the CA to constrain the upper limit of JH III production at the end of the gonadotrophic cycle.  相似文献

The calcineurin pathway has been reported to be essential for the development of azole resistance in Candida albicans. The depletion or ectopic over-expression of RTA2 increased or decreased susceptibility of C. albicans to azoles, respectively. CaCl2- induced activation of the calcineurin pathway in wildtype C. albicans promoted resistance to azoles, while the Ca 2+ chelator (EGTA), calcineurin inhibitors (FK506 and cyclosporin A) and the deletion of RTA2 blocked the resistance-promoting effects of CaCl2. Furthermore, we found that RTA2 was up-regulated in a calcineurin-dependent manner. The depletion of RTA2 also made the cell membrane of C. albicans liable to be destroyed by azoles and RTA2 over-expression attenuated the destroying effects. Finally, the disruption of RTA2 caused an increased accumulation of dihydrosphingosine (DHS), one of the two sphingolipid long-chain bases, by decreasing release of DHS. In conclusion, our findings suggest that RTA2 is involved in calcineurin-mediated azole resistance and sphingoid long-chain base release in C. albicans. Tendrils are thread-like organs whose function is to support the stems of many species of climbing plants. Tendrils naturally move (circumnutate) in space. Individual tendrils of cucumber (Cucumis sativus L.) Poinset had the vertical component of their mean velocity vector of circumnutation changed when exposed to a range of weak static magnetic fields between 1 and 16 mT. The speed (modulus) of the velocity vector was significantly increased (p=0.016) in the vicinity of a magnet, but its direction did not show a definite trend with respect to the magnet. Although cucumber tendrils bear static positive charges, they did not behave as charged bodies do in a magnetic field, neither did they show a magnetotropic response. In fact, tendrils showed a nastic response to magnetism. Magnetic fields affected some processes underlying the movement of circumnutation, but no clear interpretation of them can be given presently on the basis of the known effects of magnetism on plants. It is clear that cucumber tendrils, because of some inherent susceptibility to magnetism or their particular size and shape, are very sensitive to relatively low static magnetic field strengths.  相似文献

Summary A naturally regenerated young-growth in the Buchser Hochwald area in the Canton of St. Gallen, Switzerland, was used to determine the silviculturally permissible browsing limit for sycamore maple (Acer pseudoplatanus). The investigated area is situated at an altitude ranging from 1280 to 1310 m a.s.l. on anAbieti-Fagetum typicum site. Browsing was done by red deer (Cervus elaphus) and roe deer (Capreolus capreolus). A total of 57 sycamore maples, 1.30 m high, were examined. The actual browse impact on these plants was assessed by determining the frequency of browsing marks on the stem axis. To this end, the plants were cut into sections of 5 cm each and then split radially. On this basis it was possible to calculate permissible shares of browsed plants for four different size categories between 0.10 and 1.30 m. The permissible share of plants with two or more visible browsing marks on the stem axis amounted to 37.8% as an average value for the total risk period. This corresponds to a quota of 26.9% browsed terminal shoots per annum.  相似文献

Human bystin was identified as a cytoplasmic protein directly binding to trophinin, a cell adhesion molecule potentially involved in human embryo implantation. Although the trophinin gene is unique to mammals, the bystin gene (BYSL) is conserved across eukaryotes. Recent studies show that bystin plays a key role during the transition from silent trophectoderm to an active trophoblast upon trophinin-mediated cell adhesion. Bystin gene knockout and knockdown experiments demonstrate that bystin is essential for embryonic stem cell survival and trophectoderm development in the mouse. Furthermore, biochemical analysis of bystin in human cancer cells and mouse embryos indicates a function in ribosomal biogenesis, specifically in processing of 18S RNA in the 40S subunit. Strong evidence that BYSL is a target of c-MYC is consistent with a role for bystin in rapid protein synthesis, which is required for actively growing cells. The influence of mycorrhizal colonization withGlomus mosseae on parameters of N2 fixation and plant growth was studied in pot experiments with pea plants (Pisum sativum L.) infected withRhizobium leguminosarum and supplied with varied levels of phosphorus (P) and nitrogen (N). Reduced light intensities were used to evaluate the dependence of the microsymbionts on assimilate supply. In plants grown with low P supply, mycorrhization increased the concentration of P in shoots, and thus N2 fixation. Reduced light intensity significantly depressed mycorrhizal colonization and nodule growth in low-P plants. When P supply did not limit plant growth and N2 fixation, however, the percentage of mycorrhizal colonization was reduced due to the higher P status, and the microsymbionts were not impaired by low light intensities. To maximize carbohydrate supply, another experiment was carried out at high light intensity of 900 mol m–2s–1 and with non-limiting P supply. Nitrogen fertilization, given as starter N, enhanced plant growth, but delayed nodule formation. Towards flowering, nodulation rapidly increased, but less so inGlomus inoculated plants. After 28 days mycorrhizal plants were lower in shoot dry weight, nodule dry weight and nitrogenase activity. The results suggest that under many, but not all, environmental conditions the host plant is able to restrict mycorrhizal colonization and, thus, to prevent impairment ofRhizobium symbiosis.deceased in May 1994  相似文献
