Summary Bovine pituitary intraglandular colloid thought to be a waste product, is the holocrine secretion of intermediate lobe cells. It is housed in the intraglandular lumen (residual lumen) and is extruded into the venous circulation of the cavernous sinuses via clefts in the capsule of the gland aligned with the intraglandular lumen. Intraglandular colloid, fraction F5 (mol.wt 34,000), radiolabeled with (125I)Na and injected (0.15 ml) into the right internal jugular vein of male Wistar rats, accumulated in the endocrine pancreas. Autoradiographs showed that the material had specifically localized in the capillary network of the endocrine pancreas. Since the intermediate lobe is poorly vascularized, intraglandular colloid is considered to be the transport medium for intermediate lobe materials.This study was supported by a grant from the Natural Sciences and Engineering Research Council of Canada.I would like to thank S. Summary The production of intraglandular colloid, brought about by the cyclic breakdown of intermediate lobe cells, is related to the synthesis of hormones in the hypophysis. By analyzing the shift of the amino acid, cystine, during various phases of the gland's cycle it is apparent that hormones, as well as other protein determinants of immunological importance, are transported by way of the intraglandular cleft to the venous circulation and to the cerebrospinal fluid.This study was supported by a grant from the National Research Council of Canada.  相似文献

Recent data on the immunolocalization of regulatory peptides and related propeptide sequences in endocrine cells and tumors of the gastrointestinal tract, pancreas, lung, thyroid, pituitary (ACTH and opioids), adrenals and paraganglia have been revised and discussed. Gastrin, xenopsin, cholecystokinin (CCK), somatostatin, motilin, secretin, GIP (gastric inhibitory polypeptide), neurotensin, glicentin/glucagon-37 and PYY (peptide tyrosine tyrosine) are the main products of gastrointestinal endocrine cells; glucagon, CRF (corticotropin releasing factor), somatostatin, PP (pancreatic polypeptide) and GRF (growth hormone releasing factor), in addition to insulin, are produced in pancreatic islet cells; bombesin-related peptides are the main markers of pulmonary endocrine cells; calcitonin and CGRP (calcitonin gene-related peptide) occur in thyroid and extrathyroid C cells; ACTH and endorphins in anterior and intermediate lobe pituitary cells, alpha-MSH and CLIP (corticotropin-like intermediate lobe peptide) in intermediate lobe cells; met- and leu-enkephalins and related peptides in adrenal medullary and paraganglionic cells as well as in some gut (enterochromaffin) cells; NPY (neuropeptide Y) in adrenaline-type adrenal medullary cells, etc.. Both tissue-appropriate and tissue-inappropriate regulatory peptides are produced by endocrine tumours, with inappropriate peptides mostly produced by malignant tumours.  相似文献

Pteroyl polyglutamate hydrolase (folyl conjugase), which hydrolyses the dietary polyglutamyl folates into simple forms prior to absorption, has been shown to be induced in rat pancreas in response to dietary polyglutamyl folates but not by ingestion of synthetic unconjugated folates. Folate absorption, as measured by the rise in serum folate levels after ingestion, of dietary conjugated folates is impaired in pancreatectomised animals whereas absorption of synthetic simple folate is not. A severe build-up of folyl conjugase is observed in the lumen of control but not in pancreatectomised animals after dietary folate ingestion. These results taken together would suggest that dietary folates are hydrolysed to monoglutamyl forms suitable for absorption in the lumen; the hydrolysis is catalysed by a luminal folyl conjugase of pancreatic origin induced by dietary conjugated folates.  相似文献

In 6-day-old females ofBlattella germanica, the activity of corpora allata (CA) was inhibited in vitro by juvenile hormone III (JH III). Effective doses (281.5 and 375.4 M in the medium) were somewhat higher than (although of the same order of magnitude as) the estimated intraglandular concentration of JH III at this age, and they induced about 45% inhibition of hormonal release and a significant intraglandular accumulation of JH III and methyl farnesoate. The results suggest that autoinhibitory mechanisms operate in the CA to constrain the upper limit of JH III production at the end of the gonadotrophic cycle.  相似文献

The immunocytological reactions with anti-met-enkephalin and anti-alpha-endorphin reveal two types of fibres in the intermediate lobe of the Carp. Their pathways through the adenohypophysis and the localization of their terminals are different. The anti-met-enkephalin positive fibres terminate mainly in a zone close to the proximal pars distalis; however some fibres terminate deeper in the intermediate lobe. The anti-alpha-endorphin positive fibres penetrate mainly in the central and ventral part where thye are more numerous than the former. It seems that the anti-met-enkephalin positive fibres belong to the gomori positive preoptico-hypophyseal tract, whereas the anti-alpha-endorphin positive fibres are gomori negative.  相似文献

An immunserum was prepared from gamma endorphin and its specifity was checked by the indirect method. It was tested on hypophysis which were taken from Pigs since birth till 3 months old. The anterior lobe as well as the intermediate lobe have reacted. The same cells were positive in the anterior lobe and recognized the anti-ACTH, anti-beta MSH and anti-beta LPH immunserums. However, the reaction was more labile than with these last immunserums and it was varying with the delay of fixation after the death and the penetration in the gland by the fixator.  相似文献

Summary Dog thyroid follicular cells exposed to concanavalin A (Con A) in vitro showed changes in cell shape, induction of colloid droplets and alterations in the distribution of microvilli. Cells exposed to Con A plus suboptimal concentrations of TSH (thyroid stimulating hormone) showed pseudopods and their cytoplasm was virtually occupied with colloid droplets. The olfactory sense detects and distinguishes a multitude of different odors. Recent progress in molecular as well as physiological approaches has elucidated basic principles of neuronal encoding of odorants, common to insects and vertebrates. The construction of neuronal representations for odors begins with the task of mapping the multidimensional odor space onto the two-dimensional sensory surface, and subsequently onto the olfactory bulb or antennal lobe. A distributed expression of odorant receptors, albeit restricted to subregions of the sensory surface (large, intermediate or small for zebrafish, mouse or drosophila, respectively), ensures a robust representation, insensitive to mechanical insult. Olfactory receptor neurons expressing the same odorant receptors converge to form a receptotopic map in the olfactory bulb or antennal lobe. The emerging coding principle is a chemotopic representation of odorants at the first brain level, realized either as combinatorial or as monospecific representation, depending on the odorant.  相似文献

Elaboration and excretion of these polypeptides are enhanced after dehydration or adrenalectomy. Summary Animals having an ascites hepatoma show an increase in the incorporation of P32 into ribonucleic acid and desoxyribonucleic acid of the pancreas, whereas no change is seen in the kidneys. The variations noted in the pancreas may be compared with those observed previously in the liver and the adrenals. It can therefore be presumed that a substance which stimulates biosynthesis or disrupts the normal metabolic balance is produced by the ascites cells.

When the canine thyroid gland was stained by immunofluorescence and immunoperoxidase methods using undiluted thyroglobulin antiserum, a considerably stronger immunoreactivity was revealed in the parafollicular cells than in the colloid droplets and follicular cells. After induced hypercalcemia and antithyroid drug treatment, the immunoreactivity of the parafollicular cells coinciding with the movement of secretory granules containing calcitonin was conspicuously decreased. An application of diluted serum (above 1:10) produced a strong reaction in the colloid.  相似文献

Summary Colloid osmotic pressure (COP) of blood from the great cardiac vein was continuously measured by the use of a membrane colloid osmometer during reactive hyperemia resulting from temporary occlusion of the anterior descending branch of left coronary artery. The COP increased sharply but transiently after the release, then it decreased below the initial level and gradually recovered. It was concluded that a measurable amount of water moved from the capillary blood into the myocardial tissue and then flowed back slowly into the capillary blood.  相似文献

Autoradiographic studies using 3H-uridine in the salivary gland of adult Lygaeus sp. were carried out. The gland cell nuclei, particularly the multiple nucleoli, are the sites of incorporation of the label exhibiting RNA synthesis. The nodes formed by the mid-ventral tracheal anastomoses in abdominal segments 3-6 are modified into conspicuous glandular organs in larvae of certain Lepidoptera. They comprise clusters of extremely large cells penetrated by an extensive lacunar system opening onto the tracheal wall. These cells appear to sequester substances from the haemolymph which may be conjugated with a lipoid synthesized within them and the product excreted into the lacunar system, ultimately passing into the tracheal lumen.  相似文献

Oral administration to fasted rats of absolute ethanol produces extensive necrotic lesions of gastric mucosa as well as a massive leakage of proteins and mucus glycoproteins into gastric lumen. When the new synthetic prostaglandin MDL 646, belonging to the PGE1 series, is administered intragastrically (2 or 10 micrograms/kg) 30 min before ethanol administration, a significant protection of rat gastric mucosa against alcohol injury is observed.  相似文献

Double-labeled bile ([U. 3H glycerol] [1. 14C linoleic acid])--in which about 70% of labeling 14C and 80% of labeling 3H of total lipids were borne by phosphatidylcholines (PC), (isotopic ratio of these PC was equal to 1)--was introduced into the duodenum of test Rats, some of them with a bile fistula. As low amounts of the hydrolysis products of biliary PC were found in the intestinal lumen, a higher hydrolysis must occur further (brush border, enterocyte ?) because, in the mucosa, the highest labeling 14C was present as triglycerides and PC have an isotopic ratio 3H/14C higher than 1. As in the lumen the isotopic ratio 3H/14C of PC was higher than 1 and increased with the time elapsed, this finding suggests that mucosal PC were added to biliary PC (secretion or desquamation ?) unless these modifications were due to luminal micro-organisms. As test Rat bile was poorly labeled a very weak enterohepatic circulation of biliary diunsaturated PC may exist.  相似文献

An exceptionally efficient mechanism for the vertical transmission of a parasitic gregarine is dependent on the insect host's immune response. Gametocysts of Ascogregarina chagasi on the genital accessory glands of adult female sand flies (Lutzomyia longipalpis) become encapsulated through hemocyte-mediated immune reactions. Oocysts of A. chagasi, ejected into the lumen of the glands owing to pressure exerted by this capsule, become glued to eggshells and are subsequently ingested by larvae. In L. longipalpis with an experimentally suppressed encapsulation reaction, fewer accessory glands contained oocysts of A. chagasi.  相似文献
