Summary 3 experimentally distinct transverse relaxation components of the water in frog sciatic nerve are obtained by Carr-Purcell-Meiboom-Gill technique. The relative weights of these components: 29%; 50%; 21% fit well with water compartments in this tissue as revealed by previous methods.The previous communication in this series: V. Vasilescu, D.-G. Summary Pistia stratiotes L., a perennial water weed found in natural water bodies in Nigeria, removes organic and inorganic nutrients as it grows and spreads in the waters. In a lake which received a variety of pollutants, the plant reduced oxidizable organic matter (4-h permanganate value) by 61%, the biochemical oxygen demand by 85.3%, ammonia nitrogen by 95.3% and water soluble phosphorus by 85%. The present method makes possible a quick separation of cells from the medium with the aid of strips of filter paper and physiological solution. Cell suspension is put on the strip and all water soluble components are washed away by soaking in saline solution, while cells remain on the spot. The cell is a crowded volume, with estimated mean mass percentage of macromolecules and of water ranging from 7.5 to 45 and 55 to 92.5 %, respectively. However, the concentrations of macromolecules and water at the nanoscale within the various cell compartments are unknown. We recently developed a new approach, correlative cryo-analytical scanning transmission electron microscopy, for mapping the quantity of water within compartments previously shown to display GFP-tagged protein fluorescence on the same ultrathin cryosection. Using energy-dispersive X-ray spectrometry (EDXS), we then identified various elements (C, N, O, P, S, K, Cl, Mg) in these compartments and quantified them in mmol/l. Here, we used this new approach to quantify water and elements in the cytosol, mitochondria, condensed chromatin, nucleoplasm, and nucleolar components of control and stressed cancerous cells. The water content of the control cells was between 60 and 83 % (in the mitochondria and nucleolar fibrillar centers, respectively). Potassium was present at concentrations of 128–462 mmol/l in nucleolar fibrillar centers and condensed chromatin, respectively. The induction of nucleolar stress by treatment with a low dose of actinomycin-D to inhibit rRNA synthesis resulted in both an increase in water content and a decrease in the elements content in all cell compartments. We generated a nanoscale map of water and elements within the cell compartments, providing insight into their changes induced by nucleolar stress.  相似文献

Summary In the larval epidermal trunk region ofSalamandra maculosa, the total number of mitoses of both sides is approximately the same. This applies to the differentiated Leydig cells as well as to the rest of the epidermis cells.With respect to the meristic segmentation of the trunk, we find within the single segments a regular rise of the mitotic rate, which alternates from one side to the other.From the total number of mitoses found in the epidermis of the trunk, approximately 40% can be accounted for to the dorsal and 60% to the ventral region of the trunk. After measurement of thep H and determination of the alcalinity the cations are exchanged for H-ions in a bed of a synthetic resin exchanger. These are determined by titration with NaOH. Total and ouabain-sensitive ATPase activities were determined in the salivary glands of ticks throughout the feeding cycle. Activities were very low in unfed specimens. In the glands of feeding females, the activities rose until a maximum was reached for both ATPase components at approximately 200 mg. The activities remain low in males throughout the feeding period. These findings are discussed in relation to the fluid secretory process of the salivary glands.  相似文献

The ecophysiology of cold tolerance in many terrestrial invertebrate animals is based on water and its activity at low temperatures, affecting cell, tissue and whole organism functions. The normal body water content of invertebrates varies from 40 to 90% of their live weight, which is influenced by water in their immediate environment, especially in species with a water vapour permeable cuticle. Water gain from, or loss to, the surrounding atmosphere may affect animal survival, but under sub-zero conditions body water status becomes more critical for overwinter survival in many species. Water content influences the supercooling capacity of many insects and other arthropods. Trehalose is known to maintain membrane integrity during desiccation stress in several taxa. Dehydration affects potential ice nucleators by reducing or masking their activity and a desiccation protection strategy has been detected in some species. When water crystallises to ice in an animal it greatly influences the physiology of nearby cells, even if the cells remain unfrozen. A proportion of body water remains unfrozen in many cold hardened invertebrates when they are frozen, which allows basal metabolism to continue at a low level and aids recovery to normal function when thawing occurs. About 22% of total body water remains unfrozen from calculations using differential scanning calorimetry (compared with ca 19% in food materials). The ratio of unfrozen to frozen water components in insects is 1:4 (1:6 for foods). Such unfrozen water may aid recovery of freezing tolerant species after a freezing exposure. Rapid changes in cold hardiness of some arthropods may be brought about by subtle shifts in body water management. It is recognised that cold tolerance strategies of many invertebrates are related to desiccation resistance, and possibly to mechanisms inherent in insect diapause, but the role of water is fundamental to them all. Detailed experimental studies are needed to provide information which will allow a more complete and coherent understanding of the behaviour of water in biological systems and aid the cryopreservation of a wide range of biological material.  相似文献

A convenient one-step procedure, based upon the tyrosinase co-oxidation of dopa and cysteine, is reported for the synthesis of 5-S-cysteinyldopa (I) in 74% yield. Secondary products of the reaction turned out to be 2-S-cysteinyldopa (II, 14%), 2,5-S, S-dicysteinyldopa (iv, 5%), and the hitherto unknown 6-S-cysteinyldopa (III, approximately 1%).  相似文献

The antibodies against estrogen receptor were obtained after injecting Rabbits with a cytoplasmic receptor fraction isolated from Calf uterus. The estrogen receptor was partially proteolysed by the action of trypsin and subsequently purified by affinity chromatography (purification 4,000 to 10,000 fold, to a purity of 5-20%). The affinity of the antibody for the proteolysed receptor is KD approximately 1 nM and serum titres have reached values of approximately 50 nM. The values remained constant after the third injection. Preliminary results indicate that the antibody has approximately the same affinity for "native" cytoplasmic estrogen receptor from Calf uterus, as well as for the "trypsinized" forms of estrogen receptor isolated from Calf uterine cytosol and Hen oviduct nuclei.  相似文献

A study was made of the effect of 4 constituents of antacid preparations on the absorption of the coumarin derivate warfarin sodium using an in vitro experimental model. The constituents tested were activated dimethiocone (a silicone) magnesium trisilicate, bismuth carbonate and the adsorbant, kaolin. Slight decreased intestinal absorption was shown by magnesium trisilicate (19%) and bismuth carbonate (7%), the other 2 components showing no effects.  相似文献

The solubility of granulopoiesis inhibiting factor (GIF) in various aqueous and organic solvents was investigated. GIF is soluble in water, 10% acetic acid, and 10% pyridine. It is not extractable by 1-butanol at low and high pH. A high solubility was found in polar organic solvents (ethanol-acetone 9:1, and chloroform-methanol 1:1), whereas GIF seems to be insoluble in pure chloroform.  相似文献

A number of papers in recent years have investigated the problems of forecasting contemporaneously aggregated time series and of combining alternative forecasts of a time series. This paper considers the integration of both approaches within the example of assessing the forecasting performance of models for two of the U.K. monetary aggregates, £M3 and MO. It is found that forecasts from a time series model for aggregate £M3 are superior to aggregated forecasts from individual models fitted to either the components or counterparts of £M3 and that an even better forecast is obtained by forming a linear combination of the three alternatives. For MO, however, aggregated forecasts from its components prove superior to either the forecast from the aggregate itself or from a linear combination of the two.  相似文献

Pancreatic ductal adenocarcinoma (PDAC) is among the most devastating human malignancies, with approximately 20–30% of PDAC patients receiving the surgical resection with curative intent. Although many studies have focused on finding ideal “drug chaperones” that facilitate and/or potentiate the effects of gemcitabine (GEM) in pancreatic cancer, a significant benefit in overall survival could not be demonstrated for any of these combination therapies in PDAC. Given that pancreatic cancer is characterized by desmoplasia and the dual biological roles of stroma in pancreatic cancer, we reassess the importance of stroma in GEM-based therapeutic approaches in light of current findings. This review is focused on understanding the role of stromal components in the extrinsic resistance to GEM and whether anti-stroma therapies have a positive effect on the GEM delivery. This work contributes to the development of novel and promising combination GEM-based regimens that have achieved significant survival benefits for the patients with pancreatic cancer.  相似文献
