The antifertility action of α-chlorohydrin: metabolism by rat and boar sperm

Summary The male antifertility agent -chlorohydrin (I) is metabolized by rat and boar sperm to -chloroactaldehyde (III), -chlorolactate (IV) and Cl^– and not to the proposed active metabolite, -chlorohydrin-1-phosphate (III). It is proposed that -chlorolactaldehyde is produced intracellularly by a specific enzyme and that this is the metabolite responsible for the species-specific antifertility activity of -chlorohydrin.This work was supported by The National Health and Medical Research Council of Australia.     

Hydrolysis of amino acid β-naphthylamides by aminopeptidases in the parotid gland

Zusammenfassung Die Aktivität der Aminopeptidasen in Ohrspeicheldrüsen wurde gemessen. Glycyl-Prolin-naphthylamid, Alanin-naphthylamid, Leucin-naphthylamid, Methionin-naphthylamid, und Arginin-naphthylamid wurden von der Mikrosomenfraktion und der löslichen Fraktion schnell gespalten. Das Glycyl-Prolin-naphthylamid spaltende Enzym war in Ohrspeicheldrüsen in relativ grösserer Menge vorhanden. Die Aufspaltung von Glycyl-Prolin-naphthylamid in Glycyl-Prolin und-Naphthylamin wurde papierchromatographisch nachgewiesen.     

The microbiological dehydrogenation of 16β-methyl-5α-dehydrocortisone

Zusammenfassung Über die mikrobiologische Umwandlung des 16-Methyl-5-Dehydrocortisons in 16-Methylprednison durch einen Stamm vonCorynebacterium simplex wird berichtet. Mit Hilfe dieser Bakterienkulturen ist es möglich, in relativ kurzer Zeit die beiden Doppelbindungen in den Stellungen 1 und 4 im A-Ring des 16-Methylprednisons einzuführen.     

Resistance to β-lactam antibiotics

-lactams have a long history in the treatment of infectious diseases, though their use has been and continues to be confounded by the development of resistance in target organisms. -lactamases, particularly in Gram-negative pathogens, are a major determinant of this resistance, although alterations in the -lactam targets, the penicillin-binding proteins (PBPs), are also important, especially in Gram-positive pathogens. Mechanisms for the efflux and/or exclusion of these agents also contribute, though often in conjunction these other two. Approaches for overcoming these resistance mechanisms include the development of novel -lactamase-stable -lactams, -lactamase inhibitors to be employed with existing -lactams, -lactam compounds that bind strongly to low-affinity PBPs and agents that potentiate the activity of existing -lactams against low-affinity PBP-producing organisms.Received 9 February 2004; received after revision 30 March 2004; accepted 19 April 2004     

The in vivo expression of the globin genes of theβ cistron in γ-,δ-, andδβ-thalassemia heterozygotes

There is considerable evidence suggesting that the switch from to and chain production after birth is due, in part, to silencing of the genes by stage-specific factors which bind to their promoters and to the competition from the adult ( and ) genes for a common enhancer element located in the locus control region. As a consequence one can expect that the increased Hb F production in adults with hereditary persistence of fetal hemoglobin or -thalassemia is directed mainly by -globin genes in cis to the deletion(s) responsible for these conditions. Here we review data on heterozygotes with -, -, or -thalassemia, who also had an^AT mutation, in cis or in trans, which was used as a marker of gene expression. The results show that a deletion affecting adult genes favors the expression of genes in cis, while the deletion of a single gene does not affect the expression of the gene in cis but leads to a faster switch postnatally.     

Neue Synthesen des Chloramphenicols und deren stereochemischen Beziehungen

Summary Two new syntheses of Chloramphenicol are described starting from the diastereoisomers of -phenylserinmethyl ether and cinnamyl alcohol methyl ether respectively. The configurational correlation of the diastereoisomers of -phenylserin-methyl ether with -phenylserin and chloramphenicol is discussed. A new method for transforming erythro--phenylserinol into threo--phenylserinol is described. New examples of the Neighboring group effect were studied. The sterical course of the addition of methylhypobromite to the double bond is discussed.     

7α-hydroxylation of dehydroepiandrosterone in human testis and epididymis in vitro

Zusammenfassung Zerhackte menschliche Hoden und Epididymis wurden mit Dehydroepiandrosteron ohne Kofaktoren bebrütet und es wurden unter anderem 7-und 7-Hydroxydehydroepiandrosteron und 5-Androsten-3, 7, 17-triol charakterisiert. Die Ausbeute an 7-hydroxylierten Metaboliten war fast gleich in Hoden und Epididymis.

Summary Direct incorporation and cold trap experiments suggest that cholest-4-en-3-one is an intermediate in the conversion of cholesterol into 3-hydroxymethyl-A-nor-cholestane in the marine spongeAxinella verrucosa. Cholest-4-en-3-one is further transformed by the sponge into cholest-4-en-3-ol, 5-cholestan-3-one and 5-cholestan-3-ol; these compounds arise from side reactions, which are not part of the major metabolic route leading to 3-hydroxymethyl-A-norsteranes.     

Zur Konstitution des Resibufogenins und Artebufogenins

Summary The structure of Resibufogenin is unambiguously shown to be 3-hydroxy-14, 15-oxido-bufadien-(20, 22)-olide on the basis of new degradation work. Artebufogenin A and Artebufogenin B are 3-hydroxy-14-15-keto-bufadien-(20, 22)-olide and 3-Hydroxy-14-14-keto-bufadien-(20, 22)-olide, respectively.     

Isolement et identification du 5β-prégnane 11β, 17α, 21-triol 3, 20-dione (Dihydrocortisol) dans l'urine humaine

Summary 5-pregnane 11, 17, 21-triol 3, 20-dione (dihydrocortisol) has been isolated and identified in the urine of patients with feminizing adrenal cortical cancer. This steroid showed the same Rf as synthetic dihydrocortisol in six different chromatographic systems. After acetylation, its Rf was the same as that of 21 monoacetate of dihydrocortisol. After bismuthic or chromic oxydation, it had the same chromatographic Rf as 5-androstane 11-ol 3, 17-dione and 5-androstane 3, 11, 17-trione respectively. After chromic oxydation of its acetate, the Rf was the same as that of 21 monoacetate of cortisone.

Synthese von 5α- und 5β-Androstan-17β-ol-3-on-17-acetat aus Hyodesoxycholsäure

Summary 5- and 5-androstane-17-ol-3-one-17-acetate have been prepared from 5-androstane-6, 17-diol-3-one-17-acetate by a synthesis involving directedHuang-Minlon-reduction.Separation of 5-androstane-17-ol-3-one from its 5-isomer could readily be achieved by crystallisation of their 3-ethylenketals from acetone.     

Über die Ausscheidung von 3β-Hydroxy-17-Ketosteroiden im Harn von Meerschweinchen

Summary 11-hydroxy-epiandrosterone was isolated and identified from urine of normal untreated female guinea pigs. After cold and hot acid hydrolysis of the urine, we found another 3-hydroxy-17-oxosteroid. Its character was investigated by several methods and we suppose this steroid is9(11)-epiandrosterone, an artificial product of 11-hydroxy-epiandrosterone.

Summary The structure of Cinobufagin is shown to be 3-Hydroxy-14, 15-epoxy-16-acetoxy-5-bufa-20, 22-dienolide.

Lysosomal hydrolases in lymphoid tissue from germ-free and neonatal rats

Zusammenfassung Die Ergebnisse histochemischer Färbeverfahren für -Galaktosidase und -Glukuronidase in lymphoiden Geweben von keimfreien Ratten sind ähnlich denjenigen von neugeborenen Tieren. Diese Daten stellen einen zusätzlichen Beweis für die wichtige Rolle der Antigene bei der normalen neonatalen Entwicklung der lysosomalen Enzymaktivität dar.     

Affinity chromatography of human serum proteins using matrix bound lectin fromViscum album L.

Summary The D-galactose specific lectin fromViscum album L. reacts with serum proteins that contain the corresponding D-galactopyranosyl residues. By affinity chromatography of human serum on lectin-sepharose IgM, ₂-macroglobulin, haptoglobin and -lipoprotein were quantitatively retained. Only parts of IgA, IgG and transferrin were retarded. The other serum proteins are unbounded as albumin, ₁A– and ₁C.     

Deuterium isotope effects in the ninhydrin reaction of primary amines

Summary The rate of development of Ruhemann's purple in the ninhydrin reaction of two deuterated primary amines, -d₂-p-tyramine and -d₂--phenylethylamine, is significantly reduced It appears to be a primary isotope effect and indicates that the cleavage of the carbon-hydrogen bond at the -position is involved in the rate-determining step of the color reaction.Acknowledgements. Ein neues Verfahren zur Herstellung von 19-Norsteroiden

Summary The new methods for the production of 19-norsteroids described involve the conversion of 5-halogen-6-hydroxy-steroids into the corresponding 5-halogen-6:19-ethers either with lead tetraacetate or by the hypoiodite reaction^8,10. The 6:19-oxygen bridge is then opened reductively either directly or after oxidation of the 6:19-ethers to lactones or preferably after introduction of a ^4,6-3-oxo grouping. Acylolytic cleavage of the ⁴-3-oxo-6:19-ethers followed by alkaline hydrolysis gives ^4,6-3-oxo-19-hydroxy-steroids. The 19-hydroxycompounds formed are easily converted into 19-norsteroids by known methods.

Decrease of norepinephrine in brain and heart of vitamin E deficient rats

Zusammenfassung Viermonatiger Vitamin-E-Mangel bewirkt bei Ratten einen etwa 25 prozentigen Abfall des Noradrenalin-Gehaltes in Gehirn und Herz, der durchdl--Tokopherol rückgängig gemacht wird. Die Abnahme des Noradrenalins erfolgt ohne gleichzeitige Verminderung der Tyrosin-Hydroxylase- und Dopamin--Hydroxylase-Aktivität.
