Identification of hemolytic granules isolated from human myocardial cells

Human myocardial cells from fresh autopsy material contained granules which possessed hemolytic activity against guinea pig and rabbit erythrocytes. The hemolytic granules, which had a density of 1.02 and a diameter of 200-300 nm, were recovered as a microsome fraction from subcellular homogenates of human myocardial cells by differential centrifugation in 300 mM sucrose containing 0.1 mM PMSF and 10 mM EDTA. The membrane lesions caused by the granules were ring-like structures with an internal diameter of about 10-17 nm, analogous to that caused by perforin- and complement-induced lysis. However, the requirement for divalent cation differed from that for perforin-induced lysis, since the microsome-mediated lysis occurred in the presence of EDTA.

Summary By electron microscopic examination of a solid, chiefly scirrhous mammary cancer, 3 kinds of unusual nuclear inclusions were found: (a) Electron dense particles of a 54–80 nm diameter, whose outlines appear mainly hexagonal, which points at an icosahedral structure, (b) Clusters of granules with a diameter of 200–300å, respectively 300–400å, at whose circumference the larger particles appear, (c) Bundles of filaments in close association and continuous with the granules. In all 3 nuclear inclusions there are subunits of 40–45å. The comparison between these results and experiments published suggest that these nuclear inclusions are (a) virus particles, (b) virus at an early stage of development and (c) virus protein subunits.     

Filaments of the vertebrate lens

Summary Filaments of an average diameter of 10–12 nm have been identified in the eye lens fibre cells of representative species of each vertebrate order. Net ATP synthesis by running the red cell calcium pump backwards

Summary Ca²⁺ loaded inside-out vesicles from human red blood cells, yielding Ca²⁺ into a Ca²⁺ free medium with 4 mM EGTA, 2 mM ADP and 10 mM phosphate, produced an excess of 14.9 pmoles · min^–1 · (mg protein)^–1 of ATP compared to controls in which the transmembrane Ca²⁺ gradient was abolished by the ionophore A 23 187.We are obliged to Dr H. The hemolytic activity of heterocyclic N-alkyl amine oxides

Summary The N-alkyl derivatives of morpholine-, pyrrolidine-, piperidine- and perhydroasepine-N-oxides caused the rapid, temperature-dependent, hemolysis of human red blood cells. The most hemolytic were the amine oxides with alkyl groups having 14–18 carbon atoms.     

Exocytosis of secretory granules — a probable mechanism for the release of neuromodulators in invertebrate neuropiles

Summary Presynaptic terminals typically contain secretory granules, usually 100–200 nm in diameter, in addition to the smaller synaptic vesicles. Evidence is presented that granule exocytosis is a widespread phenomenon in invertebrate neuropiles. Such secretory release is apparently associated with morphologically unspecialized regions of the plasmalemma, rather than synaptic thickenings.Acknowledgments. Role of calcium in the histamine release induced by D-galactosamine from rat mast cells

Summary Rat peritoneal mast cells were isolated and purified by differential centrifugation in Ficoll. Cells pooled from three to four rats were suspended at approximately 10⁶ cells/ml in a buffered salt solution and incubated for 1 h at 37°C in 300 l volumes in the absence or presence (9×10^–4 M) of calcium chloride. Addition of D-galactosamine hydrochloride (DGM; 2.8×10^–4 M) caused (in addition to basal release) a mean ±SEM percent histamine release of 15.7±5.2 in the presence of Ca⁺⁺ and 19±4.9 in the absence of Ca⁺⁺ (p>0.05). It is suggested that D-galactosamine does not require extracellular Ca⁺⁺ for the release of histamine from the rat mast cell.A preliminary analysis of these results was presented at the International Symposium on calcium entry blockers and tissue protection, Rome, 15–16 March 1984.     

Inhibition of glucose transport in human erythrocytes by 2,3-dioxoindole (Isatin)

10 mM isatin (2,3-dioxoindole) inhibited glucose influx into human erythrocytes by over 30%. The inhibition is of the competitive type, where the affinity constant (K_t) was increased from 5.71 (control) to 11.11 mM in the presence of isatin with no change in V_max (130 nmol/min/ml packed cells). The observed inhibition of sugar transport by isatin was not mediated through membrane–SH groups accessible to iodoacetate, iodoacetamide, DTNB, DNP or sodium arsenite. Isatin inhibited sugar transport in the presence of 2 mM harmaline, an alkaloid inhibitor of Na⁺, K⁺–ATP_ase activity. The inhibition was non additive which suggests that these two compounds interact with the same or a similar site on the erythrocyte membrane.

Summary Primary and secondary cell walls display a beautiful fibrillar texture under the electron microscope. Microfibres of cell walls from many plants and from cultures of cellulose-forming bacteria have a remarkably constant diameter of 250–300 Å.     

Agglutination of erythrocytes using lectin-labeled spacers

Summary The mechanism of agglutination of human erythrocytes by lectin, studied using lectin-labeled gold granules as spacers (5–64 nm in size), was found to depend upon the size of the spacer but not upon the number and the mobility of the receptors.     

Somatostatin and 3-oxy-methyl-D-glucose (3-OMG) uptake in isolated chicken intestinal epithelial cells

Summary The direct effect of somatostatin on the absorption of 3-oxymethylglucose in epithelial cells isolated from the small intestine of chicken was studied. The presence of somatostatin in the incubation medium at concentrations of 3.5×10^–8 M and 7×10^–8 M produced significant dose-dependent increases in the accumulation of sugar in the enterocytes. This effect might be due to an increase in the cell membrane permeability caused by hormone action.     

Effect of enkephalins in the presence of the antibiotic bacitracin in the longitudinal muscle strip preparation from guinea-pig ileum

Summary The antibiotic bacitracin (5×10^–5–4×10^–4 M) increases the inhibition of the contractile response caused by both enkephalin release and direct application of Met-enkephalin 5×10^–7 M in the longitudinal muscle strip preparation from guinea-pig ileum. This effect is attributed to an inhibition of enkephalin degrading peptidases by bacitracin.

Summary ATPases of the amine storing granules from bovine adrenal medulla and splenic nerves are inhibited by Tl⁺⁺⁺ 3×10^–5 and 5×10^–6 M, respectively. Tl⁺ up to 10^–3 M is ineffective. By T1⁺⁺⁺ in concentrations of 10^–4 M or more, proteins are precipitated, so that the enzyme inhibiton by these concentrations is unspecific. If T1⁺ is oxidized to Tl⁺⁺⁺ in the organism, the inhibition of granular ATPase may be responsible for the alterations of the catecholamine metabolism observed in thallium intoxication.     

Effects of ouabain and furosemide on saliva secretion induced by sympathomimetic agents in isolated,perfused rat submandibular glands

Summary The presence of 10^–3 M ouabain or furosemide in the perfusate inhibited saliva secretion induced by either isoproterenol (10^–5 M) or phenylephrine (10^–5 M) from isolated rat submandibular glands and caused characteristic alterations in the electrolyte composition of saliva.     

Effects of hydralazine on the elasticity of collagen

Summary The elasticity module of collagen fibres from the tail tendon of the rat was reduced after incubation with hydralazine (10^–4 to 10^–1 mM/ml). Conduction velocity of peristaltic waves in the in vivo ureter: application of a new diameter gauge

Summary The conduction velocity of peristaltic movements of the canine ureter was measured under anaesthesia with a new type of diameter gauge using an image sensor. The peristaltic velocity was 34.1±6.2 mm/sec in 10 experiments. Noradrenaline at a low dosage of 1 g/kg i.v. reduced the resting diameter, increased the conduction velocity to 47–56 mm/sec, and approximately doubled the frequency of contraction. The application of acetylcholine also caused an increase in both frequency and conduction velocity (42–46 mm/sec). A plot of the conduction velocity against the mean period of peristaltic contraction was hyperbolic in shape.     

Caclium causes the biphasic dose-response curve for pancreatic amylase secretion

Summary High concentrations of bethanechol (10^–4 to 10^–3 M) were effective stimulants of amylase secretion from the mouse pancreas if incubations are performed in low [ca²⁺] (0.1 mM) solutions but not if normal Krebs solution (2.56 mM Ca²⁺) was used. This inhibitory effect of Ca²⁺ at high secretagogue concentrations did not appear to be mediated through the microtubules or microfilaments.     

The effect of proctolin on the adenylate and guanylate cyclases in theLocusta brain at various developmental stages

Summary Proctolin at concentrations 10^–8–10^–7 M elevated by 40% brain adenylate cyclase activity of adultLocusta migratoria migratorioides R.F. In moulting individuals, proctolin caused a decrease in brain adenylate cyclase activity, and it proved to be ineffective in the larvae. Proctolin caused only a slight decrease on guanylate cyclase activity of the brain at every developmental stage.     

Selective antiproliferative effects of thymidine

A substance with antiproliferative bioactivity in an aqueous extract ofCordyline terminalis was purified and identified by mass spectrometry to be the natural nucleoside, thymidine. 10^–5M Thymidine inhibited EL4 cell replication and decreased cell viability after 12–24 h. The effect was highly specific for this nucleoside. Treated cell cultures showed a significant increase in S phase cells and a corresponding decrease in G₁ phase cells. Nitrobenzylthioinosine (which prevented facilitated entry of thymidine) protected cells from the antiproliferative action of thymidine. A human breast cancer cell line (MCF7) was also growth-inhibited by 10^–5M thymidine but a murine lymphoma cell line (K36) was not. Thus, submillimolar thymidine has effects on cell proliferation which are selective both with respect to specificity for the compound and for different tumour cell lines.     

Prostaglandin A2 (PGA2) increases the coronary vascular resistance in the guinea-pig isolated heart preparation

Summary Prostaglandin A₂ (PGA₂) in concentrations of 1.5·10^–8 to 3·10^–6 M was found to produce concentration-dependent increase in the coronary vascular resistance of the guinea-pig isolated heart without alterations in myocardial contractile force and oxygen consumption.Supported by the Deutsche Forschungsgemeinschaft.
