Kinetics and inotropic action of probenecid in guinea-pig heart in vitro

Summary Probenecid (100–750 g·ml^–1) was found to inhibit cardiac contraction force in untreated and digoxin-treated (100 g·ml^–1) isolated right guinea-pig atrium in vitro by a reversible process, without influencing beating frequency. At low concentrations (1.5–60 g·ml^–1),¹⁴C-probenecid was accumulated into right atrium by an oxygen-dependent process. Correlation between uptake and negative inotropic action of probenecid could not be found.     

Reinforcement with naloxone of N-n-propylnorapomorphine (NPA) capability for stimulating male rat copulatory behavior

Summary Naloxone at a dose of 2 mg·kg^–1, which per se did not significantly alter the copulatory pattern of sexually active adult male rats, did significantly reduce the intromission frequency as well as latency to ejaculation when administered before a low and inactive dose of N-n-propylnorapomorphine (0.5 g·kg^–1).     

Nitric oxide-related pancreatic endocrine responses to hyperglycaemia in the conscious calf

Mean plasma insulin concentration was reduced and mean plasma glucose concentration increased following the administration of N-nitro-L-arginine methyl ester (L-NAME; 100 mol kg^–1 i.a.) in conscious calves given continuous infusions of exogenous glucose (30–60 mol min^–1 kg^–1 i.v.). It is concluded that the rise in plasma insulin concentration which occurs in these animals in response to glucose is mediated, at least in part, by a nitric oxide-related factor (NOx).     

The pharmacokinetics of VIP in dog and pig

Summary Pure porcine VIP was infused systemically in 4 conscious dogs and systemically and intraportally in 6 anesthetized pigs. At 2.3 pmoles·kg^–1·min^–1 the MCR was 10.7±1.0 ml·kg^–1·min^–1 in dog and 7.6±1.5 (systemic) and 16.5±2.0 (portal) in pig. The t 1/2's were 1.0±0.12 and 1.0±0.05 respectively. These values agree with those observed in man. This very high single pass tissue clearance does not suggest a hormonal role for VIP.     

Permeability of arthrodial membrane to water: A first measurement using in vivo techniques

Summary Despite differences in surface morphology and fine structure, the permeabilities of untreated scorpion sternite and pleural cuticle to water are similar (0.69 versus 0.79 g·cm^–2·h^–1·mm Hg^–1). Hexane applied to pleural membrane increased its permeability 9-fold, but neither hexane nor chloroform: methanol had much effect on sternite permeability. When sternite cuticle was treated with 1.0 N KOH followed by chloroform: methanol, permeability increased about three times over control values. In contrast, cockroach pronotum, which is 17 times more permeable than scorpion sternite, exhibited a marked increase in permeability when treated with just hexane. In both the scorpion pleuron and cockroach pronotum, disruption of the lipid barrier caused by rubbing is partially responsible for the higher permeabilities observed following solvent treatment.     

Central nicotinic receptor blockade inhibits emotionally conditioned pressor responses in rats

A conditioned stimulus previously paired with electric footshock produced an increase in blood pressure in conscious, freely moving rats. The conditioned pressor response was reproducible. Intracerebroventricular injection of the nicotinic receptor antagonists hexamethonium (1–10 g) or pentolinium (10 g) but not the muscarinic receptor antagonist methylatropine (3 g) produced an inhibition of the conditioned pressor response, whereas intraarterial injection of hexamethonium (10 g) did not affect the response. Intraventricular injection of the cholinesterase inhibitor physostigmine (3–10 g) produced an enhancement of the conditioned pressor response. These results are consistent with the possibility that central nicotinic receptors play a role in the expression of the emotionally conditioned pressor response in rats.     

The action of iodoacetate on the antigenic power of insulin

Résumé Nous avons observé que le iodoacétate en concentrations de 1·10^–4, 5 · 10^–5 et 1 · 10^–5 M et préalablement incubé à 37 °C, a une influence de l'ordre de 61, 50 et 44% respectivement sur la totalité du pouvoir antigénique de l'insuline de buf. L'effet inhibitif est proportionnel à la durée d'incubation et l'on ne constate pas de différences lorsque les concentrations de l'hormone se trouvent entre 100 et 300U/ml.     

Drug uptake by lung slices from paraquat-pretreated rats

Summary Lung slices from male Sprague-Dawley rats pretreated with paraquat (PQ) (100 moles·kg^–1, i.v.) 16 h before sacrifice, accumulated less PQ in vitro than lung slices from saline-treated controls. Neither lung slices from PQ-pretreated nor saline control animals released (effluxed) PQ accumulated in vitro. The accumulation and efflux of imipramine and 5-hydroxytryptamine by lung slices was unaffected by prior in vivo administration of PQ.     

Choleretic and cholestatic effects of infused bile salts in the rat

Summary In rats, at low infusion rates taurocholate (TC), taurochenodeoxycholate (TCDC) and taurodeoxycholate (TCD) each produced an increase in bile flow of 20–50%. However, at high infusion rates (5–20 moles min^–1 kg^–1) the cholestatic effects of the bile salts were revealed and the relative toxicity of the bile salts was seen to be TDC>TCDC>TC.     

Hemodynamic studies in a parabiotic model of portal hypertension

Summary Splanchnic and systemic hemodynamic studies were performed in a rat model of parabiosis and portal hypertension. A portal hypertensive and a normal rat were surgically united side to side. A hyperdynamic circulation, characterized by increased cardiac index (413±26 vs 318±23 ml·min^–1·kg^–1; p<0.05) and portal venous inflow (9.61±1.29 vs 6.33±0.36 ml·min^–1·100 g b.wt^–1; p<0.05), was found in all the portal hypertensive rats but not in the normal parabiotic partners. These results do not support the existence of a transferable humoral factor mediating the hyperdynamic circulatory state of chronic portal hypertension.Acknowledgment. Cleavage of des-Arg9-bradykinin by angiotensin I-converting enzyme from pig kidney cortex

Summary Fast and very slow hydrolyses of des-Arg⁹-bradykinin and angiotensin II by angiotensin I-converting enzyme were detected by high performance liquid chromatography. The Michaelis constants of the enzyme, K_m values, for des-Arg⁹-bradykinin and bradykinin were found to be 0.24 mM and 4.4 M, and the maximum velocities, V_max values (mol·min^–1·mg protein^–1) for these compounds to be 3.24 and 0.34, respectively. The enzyme also hydrolyzed Z-Gly-Pro-Gly-Gly-Pro-Ala to a tripeptide that was identified as dansyl-Gly-Pro-Ala by TLC on polyamide. These observations show that the enzyme hydrolyzes the peptides at the bond before the prolyl residue in the penultimate position.Acknowledgments. The effect of small doses of antidiuretic hormone on renal excretion of sodium and water in saline loaded acutely hypophysectomized rats

Summary Small doses of vasopressin (200 U min^–1 kg^–1) did not influence the loss of ability of acutely hypophysectomized rats to react by natriuresis to the extracellular fluid volume expansion with saline, inspite of restoring their arterial blood pressure to normal. It is concluded that the level of antidiuretic hormone efficient enough to concentrate urine is not a decisive factor in the homeostatic mechanism promoting the volume natriuresis.Acknowledgment. Intracellular inclusions associated with maize mosaic virus infection

Zusammenfassung Die Epidermiszellen von mit Mais-Mosaik-Virus infizierten Maisblättern enthalten runde oder längliche Einschlusskörper, welche meist dem Kern anhaften oder nahe bei diesem liegen. Sie erscheinen nach Giemsa-Färbung rosa bis purpurn gefärbt. Die Abmessungen der länglichen Einschlüsse betragen 3,0–4,6 · 14–26, der Durchmesser der rundlichen 2,0–7,7.     

Dissociation-constants of metal-ion-complexes with alkaline phosphatase from pig kidney

Summary Using metal-ion buffers it was possible to remove Zn²⁺, Mg²⁺ and Mn²⁺ ions of pig kidney alkaline phosphatase reversibly. The dissociation constants obtained are K_EMg:4·10^–7 M, K_EMn:4·10^–8 M and K_EZn:8·10^–13 M (22°C, pH:9.6, :0.07).Acknowledgement: The authors thank Dr.H. U. Soybean photosynthesis: simultaneous14CO2 and O2 estimates

Summary A method of estimating the photosynthetic rate of soybean leaves using an oxygen electrode is presented. The procedure is rapid, requires small samples and compares favourably with estimates by other techniques. Light saturation occurs at 1200 E·m^–2·sec^–1. The apparent K_m for HCO ₃ ^– is 3.2 mM at pH 7.6.     

Evidence against a role of (Na++K+-ATPase in the alpha-adrenoceptor mediated positive inotropic effect of phenylephrine

Summary Phenylephrine (0.1–100 M) in the presence of 1 M propranolol increased the force of contraction in electrically driven papillary muscles from cats. This presumably alpha-adrenoceptor mediated positive inotropic effect of phenylephrine occurred without any influence on (Na⁺+K⁺-ATPase activity.This work was supported by the Deutsche Forschungsgemeinschaft.     

D-ala2-Metenkephalinamide blocks the synaptically elicited cortical spreading depression in rats

Summary Spreading depression (SD) was elicited in rats anesthetized with pentobarbital by a train of 8 electrical pulses (0.1 ms, 10 Hz) applied to parietal cortex. Local application of 50 g of D-ala²-metenkephalinamide (DAME) on the stimulated area evoked one or two SD waves followed by an increase of SD threshold from 40 V to 90 V. This effect could be partly prevented by naloxone (1 mg/kg i.p.) and reversed by local application of 4-aminopyridine (10^–3 M, 2 l), which reduced SD threshold to 5 and 20 V in normal and DAME-treated cortex, respectively. It is argued that DAME exerts an inhibitory effect on cortical neurons and that the initial SD facilitation is due to initial blockade of inhibitory neurons in the superficial cortical layers.supported by the European Training Program in Brain and Behavior Research.     

Cartsian diver studies on respiration of rat bone marrow cells in the presence of erythropoietin and anti-erythropoientin

Résumé L'étude de la respiration des cellules de la moelle des os du rat fut faite avec un plongeur Cartésien. Ces cellules, avec ou sans glucose, repirent à raison de 0,73±0,15 et 0,97±0,17×10^–6 l O₂/cellule/h. Comparée aux mesures de contrôle, la respiration des cellules médullaires de rats pléthoriques est plus fabile en présence de ESF (0,45 contre 0,99×10^–6 l, O₂/cellule/h). Cette inhibition fut supprimée par la présence d'anti-ESF (0,98 contre 0,99×10^–6 l O₂/cellule/h).

Effects of cerebral lateral,ventricular infusions of phloridzin on feeding and body weight in Gallus domesticus (L.)

Summary Contrary to earlier findings in rats, cerebral lateral ventricular infusions of 1×10^–3 M or 2×10^–3 M solutions of phloridzin at a rate of 2.5 l/min for 90 min had no significant stimulating effects on food intake and weight gain in hens and cocks. These different responses to intraventricular phloridzin might reflect a difference of sensitivity to the inhibitory action of phloridzin on glucose transport in cerebral cells or certain peculiarities of mechanisms controlling food intake in chickens.     

Effects of selective dopamine D1 and D2 antagonists on male rat sexual behavior

Summary The effects of selective dopamine (DA) D₁ and D₂ antagonists on male rat sexual behavior were investigated. The D₁ antagonist (+)SCH-23390, 25–100 g kg^–1 s.c. –20 min, and the D₂ antagonist raclopride, 0.1–1.6 mg kg^–1 s.c., –20 min, decreased both the number of mounts and intromissions preceding ejaculation. No statistically significant effects in the time up to ejaculation or in the time up to the first intromission were noted, whereas both compounds produced a statistically significant increase in the post-ejaculatory interval. The effect can generally be characterized as psychomotor inhibition, and no evidence was obtained for a specific role of DA D₁ or D₂ receptors in the mediation of male rat sexual behavior.The expert technical assistance of Ms Elisabeth Wallin is gratefully acknowledged.
