We employed in vitro translation of mRNA and product separation using SDS-PAGE to examine the heat-shock response of the worker honey bee. Increases in the levels of 6 translatable RNA populations were observed following heat stress. The greatest response was observed among bees aged 9 days. Kinetics of BRCA1 regulation in response to UVC radiation   总被引:1,自引:0,他引:1  
To investigate changes in BRCA1 following DNA damage, we exposed MCF-7 cells to increasing doses of ultraviolet C. We observed an increase in BRCA1 protein levels above 78 J/m2. This increase was observed as early as 5 min after irradiation. BRCA1 levels were then observed to decrease after 2 h, consistent with the previously published data. By pretreating with cycloheximide prior to irradiation, we observed a decrease in the protein half-life, from 3.5 h to 53 min, suggesting that a decrease in protein half-life may cause the lower levels of BRCA1 after irradiation. We also observed an increase in BRCA1 mRNA within 15 min of irradiation, followed by a decrease after 4 h. These data suggest that newly translated protein may contribute to increases in BRCA1 protein levels. The very rapid changes in BRCA1 support its role as a sensor of DNA damage, as opposed to being a repair gene. Aminopeptidase activities, assayed as arylamidase activities, were investigated in selected tissues of 1, 6, 12 and 24-month-old rats. The enzyme activities were found to have a heterogeneous distribution and age-related changes were observed. The highest levels of soluble arginyl-aminopeptidase activity were detected in brain homogenate at all the studied ages, whereas membrane-bound activity presented the highest levels in brain and kidney in the four ages tested. Aspartyl-aminopeptidase activity was detected mainly in the particulate fraction of kidney at all four ages. In 1, 6 and 12-month-old animals, soluble aspartyl-aminopeptidase activity was also higher in the kidney than in the rest of the tissues, whereas in the group of 2-year-old rats, the highest levels were found in both kidney and liver. Age-related changes were observed in all the studied tissues and for all the assayed enzymatic activities. In general, the maximal levels were detected in both the youngest and the oldest animals, and the minimal ones in 6 and 12-month-old rats. However, in the adrenals, the soluble and membrane-bound arginyl-aminopeptidase activity was higher in 6-month and 2-year-old rats than in 1-month and 12-month-old rats. These changes may reflect the functional status of the susceptible endogenous substrates of aminopeptidases.  相似文献

Levels of cortisol, estradiol and progesterone were measured in 172 amniotic fluid samples collected by amniocentesis during late pregnancy. An increase in cortisol and estradiol levels was observed three weeks before vaginal delivery of term and premature infants; in both types of delivery, onset of labor was spontaneous. In contrast progesterone levels were only slightly decreased in these cases.  相似文献

Summary Orally administered ethanol produced dose-related suppression of IRI levels and elevation of IRG levels in fed and fasting rats. The increment was not uniform thoughout the central nervous system. Particularly significant were the 25% and 80% increases observed, respectively, in whole brain and spinal cord. The levels of dopamine and norepinephrine remained unchanged. The subsequent studies of 5-HT turnover revealed a synthesis rate in the tetanic animals that was almost double that of controls. The degradation rate of the amine as well as the levels of 5-hydroxyindolacetic acid were unaffected.  相似文献

Summary Intact and TPTx animals showed the expected afternoon increase in serum LH, FSH and prolactin levels. But the afternoon increase in serum LH levels in TPTx rats was less than that observed for intact animals (p<0.01). The effect of clofibrate on rat liver phospholipid biosynthesis was studied using 32P as a precursor. Phospholipid classes, levels and specific radioactivity were evaluated. Significant increases in levels of phosphatidylethanolamine and phosphatidylcholine were found and could account for the observed increase in total phospholipids. Specific activity of phosphatidylserine increased and that of phosphatidylethanolamine decreased. This fact suggests that clofibrate seems to alter the systems engaged in the transformation occurring within the different classes of phospholipids but not the de novo biosynthesis.  相似文献

The levels of GSH-related enzyme activities during pregnancy were determined. A significant increase in Selenium-dependent GSH peroxidase and GSH S-transferase E activity was observed. Summary 3 auxin protectors, which were o-dihydroxyphenols of high molecular weight, were isolated fromZizyphus gall tissues. An increase in polyphenol oxidase activity in the gall tissue was observed which probably led to the production of high levels of auxin protectors. Summary Methanobacterium thermoautotrophicum, Methanococcus vannielii andMethanobrevibacter arboriphilus, the 3 methanogenic bacteria tested, possessed an ornithine acetyltransferase and an arginine-sensitiveN-acetylglutamate 5-phopshtransferase. These 2 enzymes, as well as acetylornithinase and ornithine carbamoyltransferase, were insensitive to oxygen and were present at activity levels comparable to the levels observed in eubacteria.Acknowledgments.
