Synthesis of L-4-amino-3-isoxazolidinone,the unnatural stereoisomer of cycloserine and its antibiotic activity

Zusammenfassung Eine direkte Synthese vonl- undd-Cycloserin ausl- undd-Serin wurde beschrieben. Die bakteriostatischen Effekte der drei Formen von Cycloserin (d-,l-unddl-) aufE. coli in synthetischem Medium wurden verglichen.

Summary Demethylation ofl- andd-adrenaline was studied on narcotized catsin vivo.In metabolism of infusedl-adrenaline, formation of noradrenaline represents one first step.On the other hand, metabolism ofd-adrenaline does not show any intermediate formation of noradrenaline.     

Curamycin. II. Structure of the hydrolysis products ‘curacin’ and ‘d-curamicose’

Zusammenfassung Curacin, die chlorhaltige Fraktion der milden Hydrolyse des Antibiotikums Curamycin, wurde durch ihre chemischen und physikalischen Eigenschaften als 4-O-Dichlor-iso-everninyl-2-deoxy-d-rhamnose aufgeklärt. Ausserl-Lyxose und 4-O-Methyl-d-fucose besitzt auch das Curamycin ein drittes Monosaccharid,d-Curamicose, welches als 2,6-Di-O-methyl-d-mannose identifiziert wurde.     

The chemistry of the polymyxin antibiotics

Zusammenfassung Die Chemie der Polymyxine, einer Klasse von basischen Polypeptid-Antibiotika, begann 1954, als durch Gegenstromverteilung erstmals ein definierter Vertreter, das Polymyxin B₁ in reiner Form isoliert werden konnte (L. C.Craig). Partialhydrolyse mit Mineralsäuren führte zum Schluss, dass es sich um Cyclohepta- oder Cyclooctapeptide mit Seitenketten handelt, die, -Diaminobuttersäure (Dab) enthalten (W.Hausmann).Amidartige Verknüpfung der Seitenkette mit einer Fettsäure [(+)-6-Methyloctansäure (MOA) oder 6-Methylheptansäure (IOA)] (S.Wilkinson) verleiht diesen Antibiotika den Charakter von Invertseifen. Sie sind besonders gegen gramnegative Erreger wirksam. Schwierigkeiten bereiteten die Aufklärung der Verknüpfungsweise der Seitenkette (- oder-) und die Beantwortung der Frage, ob das Molekül nebend-Phenylalanin in der Ringsequenz noch einend-, -Diaminobuttersäurerest in Nachbarschaft zur Fettsäure in der Seitenkette enthält. Synthetische Versuche mitd-, -Diaminobuttersäure an dieser Stelle führten zu hochaktiven Produkten, die aber mit natürlichem Polymyxin B₁ nicht identisch waren. Entscheidende Fortschritte wurden mit dem bakteriellen Enzym Nagarse (T.Suzuki) erzielt, das schrittweise die Seitenkette bis zum Ringpeptid abbaut. Dabei ergab sich, dass den Polymyxinen die allgemeine Struktur eines Cycloheptapeptides mit-verknüpfter Seitenkette zukommt (Figur 4).Die Polymyxine B₁ E₁ (Colistin A) sowie Circulin A unterscheiden sich voneinander nur durch eine Variation in der gleichen Dipeptidsequenz des 7-gliedrigen Ringes. Die im Polymyxin B₁ vorhandene Dipeptidsequenzd-Phe-l-Leu ist in Polymyxin E₁ (Colistin A) durchd-Leu-l-Leu und in Circulin A durchd-Leu-Ll-Ile ersetzt. Im Polymyxin D₁ ist neben dem Ersatz der entsprechenden Sequenz durchl-Leu-l-Thr noch ein, -Diaminobuttersäurerest der Seitenkette durch eind-Serin ausgetauscht. Die entsprechenden Verbindungen mit dem Index 2 unterscheiden sich von denjenigen mit dem Index 1 durch einen Austausch der (+)-6-Methyloctansäure durch 6-Methylheptansäure.Die Struktur von Polymyxin B₁ E₁ und Circulin A konnte durch Totalsynthese gesichert werden (K.Vogler). Weitere Fortschritte in der Erforschung der Natur der noch unbekannten Vertreter sind in Kürze zu erwarten.

Zusammenfassung Es wird über den Einfluss von einigend-Aminosäuren auf den morphologischen Charakter vonFusarium roseum Link. berichtet. Von den geprüften Aminosäuren spieltd-Arginin eine wichtige Rolle für die Bildung der hefeähnlichen Zellen.l- und besondersd-Valin beschleunigen die Bildung von Chlamydosporen-ähnlichen Zellen.     

Ein neues Alkaloid aus dem Mycel einesClaviceps purpurea-Stammes. Zur Biogenese von Ergocristin

Summary A new alkaloid from the lyophilized mycelia of aClaviceps purpurea strain, N-[N-(d-Lysergyl)-l-valyl]-l-phenylalanyl-d-prolin-lactam, has been isolated and characterized. From its fast methanolysis it is deduced that the previously reported N-(d-Lysergyl)-l-valinmethylester is actually an artefact. The biogenetic significance and its possible role in biogenesis of Ergocristine is discussed.

Effets d'analogues du chloramphénicol sur la différenciation de l'∄uf de l'oursinParacentrotus lividus

Summary The effects ofl-threo-chloramphenicol and thiophenicol on the differentiation of sea-urchin eggs are compared with those exerted byd-threo-chloramphenicol. Onlyd-threo-chloramphenicol caused vegetalization;l-threo-chloramphenicol inhibited development. These effects resemble those of nitrobenzene. Thiophenicol, differing fromd-threo-chloramphenicol by replacing the nitro group with a methylsulphonyl group, slows down development at high concentrations. The effects of chloramphenicol on oxidative phosphorylation and synthesis of proteins are discussed. It is suggested that both the steric configuration typed-threo of the side chain and the presence of a nitro group are necessary for the vegetalizing activity.     

Bildung eines ascorbigenähnlichen Produktes ausp-Hydroxybenzylalkohol und L-Ascorbinsäure

Summary l-Ascorbic acid reacts withp-hydroxybenzyl alcohol in a similar way as with 3-hydroxymethyl indole in the formation of ascorbigen (bound ascorbic acid). The crystalline reaction product was identified as a 2-C-(p-hydroxybenzyl)-3-keto-hexulosonic acid lactone.

Regionale Inkorporation der optischen Isomeren der Aminosäure Prolin-H3 im Autoradiogramm des Mäusegehirns

Summary Comparative labelling of the mouse brain with tritiatedd, l-proline andl-proline reveals a dissimilarity of incorporation between the optical isomers of this amino acid. The differential regional distribution pattern ofd-proline can be correlated to the cerebral distribution of the enzyme activity ofd-aminoacid-oxidase in the mouse brain.

Effect ofl- andd-thyroxine on the supraoptic neurosecretory ganglion cells of the rat

Zusammenfassung Mittels grosser Dosen vonl- undd-Thyroxin erhielt man keine deutlichen Veränderungen in der Menge und Verteilung von neurosekretorischem Material in den Zellen des Nucleus supraopticus bei der männlichen Ratte. Das Volumen der Kerne dieser Zellen nahm dagegen zu, besonders bei mitl-Thyroxin behandelten Ratten. Diese Veränderungen sprechen für eine vermehrte neurosekretorische Aktivität dieser Zellen.

Interaction between sugars and amino acids in intestinal absorption by rat,in vivo

Summary The inhibitory action ofl-leucine on the intestinal absorption ofd-glucose andd-galactose, as well as the inhibitory action ofd-galactose on the absorption ofl-leucine at various concentrations by rat small intestine has been studied. The further effect was more clearly evidenced when the medium was perfunded through the intestine in a closed circuit system using a peristaltic pump.     

Erythropoietic response to isomeres of triiodothyronine

Zusammenfassung Verabreichung vond-, wie auch vonl-Triiodothyrosin erzeugt eine ähnliche Vermehrung der Erythropoiesis in nephroektomierten Ratten, wie bei der Messung der Radioeiseninkorporation durch Erythrozyten (18 h nach Injektion von radioaktivem Eisencitrat). Es ergab sich weiter, dassd- undl-Thyrosin, im Unterschied zu Erythropoieten, die reticulozytose Eisenassimilation in vitro vermehrte. Danach scheinen verschiedene Mechanismen bei der Stimulation der Erythropoiesis durch Erythropoieten und Triiodothyronin im Spiele zu sein.     

Kompensation der teratogenen Wirkung eines Thalidomid-Metaboliten durchl-Glutaminsäure

Summary Skeletal defects were observed in the off-spring of SWS-mice following i.p. administration of phthaloyl-l-glutamic acid, a metabolite of thalidomide. The fetuses of animals which receivedl-glutamic acid in addition to the teratogenic drug, did not show any malformations, whereas concurrent i.p. administration of thed-isomer of glutamic acid failed to reverse teratogenicity induced by phthaloyl-l-glutamic acid.     

Avilamycin

Summary A strain ofStreptomyces viridochromogenes produced a new crystalline antibiotic, Avilamycin, related to but not identical with Curamycin and Exfoliatin. Avilamycin, C₆₃H₉₄O₃₅Cl₂, gave on solvolytic degradation the following products: dichloroisoeverninic acid, 2-deoxy-d-rhamnose, 2,6-di-O-methylmannose, 4-O-methylfucose,l-lyxose and 3,5-diacetoxy--caprolactone.     

Catalytic properties of synthetic pentapeptides

Zusammenfassung Es wurde das Pentapeptidl-Thr-l-Ala-l-Abu-l-His-l-Asp synthetisiert, seine katalytische-hydrolytische Wirkung auf Essigsäure-p-nitrophenylester geprÜft und die katalytische Aktivität ca. 1/3 derjenigen seines isosterenl-Thr-l-Ala-l-Cys-l-His-l-Asp festgestellt.

Zusammenfassung Die Strukturaufklärung von zwei neuen tryptophanhaltigen Peptiden im Schlangengift vonAgkistrodon halys blomhoffii ergeben:l-Pyroglutamyl-l-Glutaminyl-l-Tryptophan undl-Pyroglutamyl-l-Asparaginyl-l-Tryptophan. Diese Peptide sind in Schlangengiften der Viperidae- und Crotalidae-Arten verbreitet.

Über den Einbau von Dipeptiden in Ergotoxin-Alkaloide

Summary l-Valyl-l-leucine-U-¹⁴C,l-valyl-l-proline-U-¹⁴C-lactam andl-leucyl-l-proline-U-¹⁴C-lactam were administered to submerged cultures of an ergotoxine-mixture-producing strain ofClaviceps purpurea. The added dipeptides were split by the fungus into the corresponding amino acids prior to incorporation. Apparently dipeptides are no immediate precursors for the peptide side-chain of ergotoxinealkaloids.
