The influence of N2-fixation on the carbon balance of leguminous plants   总被引:3,自引:0,他引:3  
Biological dinitrogen fixation by legume-rhizobia symbiosis is very important both from the economic and from the ecological point of view. Theoretically, the reduction of the N2-molecule to ammonia requires at least 16 ATP and 1.5 mg C per mg N fixed (Nfix). These values are difficult to determine in situ as this necessitates the determination of that part of root respiration which drives N2-fixation. New approaches to such determinations and the results obtained are described. The values vary, depending on the plant species studied, the developmental stage of the plants and the genetic variability of macro- (and micro-?) symbionts. The values range between 1.5 and 4 mg C/mg Nfix. In some species (e.g.Vicia faba L. cv. Fribo), the apparent CO2 assimilation is enhanced in order to meet this high energy need. In others (e.g.Pisum sativum L. cv. Grapis), root growth is restricted. Physiological criteria are discussed which allow an early diagnosis of the energetic efficiency of various combinations of macro-and microsymbionts as a basis for a selection in plant breeding.  相似文献

After a short summary on the ecology and rhizosphere biology of symbiotic bacteria and vesicular-arbuscular (VA) mycorrhiza fungi and their application as microbial inocula, results on competitiveness and communication are summarized. Stress factors such as high temperature, low soil pH, aluminium concentrations and phytoalexins produced by the host plants were studied withRhizobium leguminosarum bv.phaseoli andRhizobium tropici onPhaseolus beans. Quantitative data for competitiveness were obtained by usinggus + (glucoronidase) labelled strains, which produce blue-coloured nodules. ForPhaseolus-nodulating rhizobia, a group specific DNA probe was also developed, which did not hybridize with more than 20 other common soil and rhizosphere bacteria. Results from several laboratories contributing to knowledge of signal exchange and communication in theRhizobium/Bradyrhizobium legume system are summarized in a new scheme, including also defense reactions at the early stages of legume nodule initiation. Stimulating effects of flavonoids on germination and growth of VA mycorrhiza fungi were also found. A constitutive antifungal compound in pea roots, -isoxazolinonyl-alanine, was characterized.  相似文献

TheRhizobium meliloti C4-dicarboxylate transport (Dct) system is essential for an effective symbiosis with alfalfa plants. C4-dicarboxylates are the major carbon source taken up by bacteroids. Genetic analysis of Dct mutant strains led to the isolation of thedct carrier genedctA and the regulatory genesdctB anddctD. The carrier genedctA is regulated in free-living cells by the alternative sigma factor RpoN and the two-component regulatory system DctB/D. In addition, DctA is involved in its own regulation, possibly by interacting with DctB. In bacteroids, besides the DctB/DctD system an additional symbiotic activator is thought to be involved indctA expression. Further regulation ofdctA in the free-living state is reflected by diauxic growth of rhizobia, with succinate being the preferred carbon source. The tight coupling of C4-dicarboxylate transport and nitrogen fixation is revealed by a reduced level of C4-dicarboxylate transport in nitrogenase negative bacteroids.  相似文献

Summary A container system was constructed to study the response of soybean roots to infection by mycorrhizal or pathogenic fungi. The system allows a rapid and synchronous inoculation byGlomus mosseae orRhizoctonia solani. The phytoalexin glyceollin was measured in roots of inoculated and uninoculated plants for a period of 30 days. A significantly increased content of phytoalexin was found inR. solani-infected roots as compared to uninfected control roots. However, there was no difference in the glyceollin contents of the mycorrhizal and the control roots for up to 23 days after inoculation. The accumulation of glyceollin inR. solani-infected roots was not influenced by a subsequent inoculation withG. mosseae. Moreover glyceollin accumulated in mycorrhizal plants to the same extent as in control plants when they were inoculated withR. solani. The two fungi did not mutually influence the course of infection when they were inoculated together.  相似文献

Summary In transfilter associations ofPortulaca grandiflora callus tissues withRhizobium sp. cowpea 32H1 the nitrogenium fixed by the bacteria was channelled into the normal pathway of nitrogen metabolism of higher plants. In associations kept in an15N2 containing atmosphere 10% of the15N taken up into the plant cells was incorporated intoPortulaca proteins. The results demonstrate that cells of nonleguminous plants are profiting from the quasi-symbiotic situation.  相似文献

Waste water purification is characterized by intensive mineralization and nitrification processes. Because of the high O2 demand, temporarily anaerobic conditions may be produced, and denitrification by nitrifying organisms as well as heterotropic denitrification may contribute to N2O release. In situ measurements (1993–1994) suggest that N2O is released from activated sludge in a domestic waste water treatment plant at an average rate of 1040 g m–2h–1 with a range between zero and 6198 g m–2h–1. The production of N2O seems to be related to the concentration of NO 2 and NO 3 as well as to the pH. In the waste water about 75–200 g N2O l–1 is dissolved. This N2O is released after discharge into the receiving waters. The N2O is produced essentially by nitrification rather than by heterotropic denitrification. On a long-term scale the increasing use of mechanical-biological waste water purification plants world-wide may add increasingly to the anthropogenic production of N2O, although the present amount of N2O produced is negligible compared to its global terrestrial production.  相似文献

Several 4-(aminomethylisoxazolyl)-1,2-naphthoquinones inhibited growth and DNA synthesis inTrypanosoma cruzi and stimulated O2 uptake and generation by the parasite epimastigotes and their mitochondrial and microsomal membranes; these results support the idea that oxygen radicals play a role in quinone toxicity. Maximal effects on respiration and generation were observed with antimycin-inhibited cells. Similar results as well as stimulation of H2O2 production were obtained withCrithidia fasciculata despite the presence of catalase in this organism.Acknowledgments. The question of whether ammonium uptake influences the occurrence of ureides in legumes has been addressed in this study by investigating threeP. vulgaris genotypes as well as one cultivar ofGlycine max. All plants were raised in sand culture during the dry season in northern Thailand and irrigated daily with nitrogen-free nutrient solution, or the same solution containing 12 mol m–3 nitrogen in the form of (NH4)2SO4 or KNO3, each treatment consisting of different proportions of either compound. Regression analyses of xylem sap composition relative to ammonium vs. nitrate supply of plants harvested at V4, R1 and R6 indicated close positive correlations of xylem amino nitrogen content and negative correlations with xylem nitrate content and ammonium supply. Statistically significant correlations between relative xylem ureide content and ammonium availability could be established for theP. vulgaris cultivar Brilliant up to stage R1, but not for the other plants investigated. It was concluded that at least for some genotypes of common bean a relationship exists between ureide production and ammonium uptake by the root system. Since the extent to which ureide production is stimulated remains quite small, its relevance to the xylem solute technique for measurement of N2 fixation may be limited. Nevertheless, due to the possibility of large genotypic differences in the impact of ammonium on ureide production, this factor must be considered in calculations if N2 fixation is to be determined in soils containing significant amounts of ammonium, e.g. in paddy fields.  相似文献

This communication compares some biochemical methods for quantifying colonization by arbuscular mycorrhizal (AM) fungi. The degree of mycorrhizal colonization can conveniently be measured by determining fungal specific sterols. AM-colonized plants show a specific synthesis of 24-methylene cholesterol and an enhanced level of campesterol (=24-methyl cholesterol). A gene probe for nitrate reductase, the key enzyme for nitrogen assimilation, has been developed, which allows the monitoring of the distribution of this enzyme in fungi. Among the phytohormones tested, only abscisic acid (ABA) is found at a considerably higher level in AM-colonized plants than in controls. The concentration of ABA is about twenty times higher in spores and hyphae of the AM fungusGlomus than in maize roots. Other phytohormones (auxins, cytokinins) do not show such alterations after mycorrhizal colonization. The roots of gramineous plants become yellow as a result of mycorrhizal colonization. The yellow pigment(s) formed is (are) deposited in larger quantities in the vacuole(s) of the root parenchyma and endodermis cells during the development of the gramineous plants. A substance isolated from such roots has now been identified as a C-14 carotenoid with two carboxylic groups, and named mycorradicin.  相似文献

Summary The herbicide paraquat (1,1-dimethyl-4,4-bipyridylium ion) has been found to exert a growth inhibitory effect on the N2-fixing blue-green algaNostoc muscorum in nitrogen-free (N2) and NO 3 media, without any apparent inhibitory or stimulatory effect on the heterocyst-forming capacity of the organism. With a dose of paraquat permitting about 20 and 50% survival of this alga a reverse mutation (fromhet + nif auxotrophy tohet + nif + prototrophy), a forward mutation (for L-methionine-DL-sulfoximine [MSO]-resistance), and an auxotrophic mutation (for carbon-auxotrophy through methylamine [MA]-resistance) have been obtained. The toxic and mutagenic effects of this agrochemical have been compared with those of the well known mutagen MNNG (N-methyl-N-nitro-N-nitrosoguanidine) and found to be stronger than those of the latter in each case.The author wishes to thank Mr Girish Nath, Lakshami Chemicals Pvt. Summary To investigate the roles of adenosine A1 and A2 receptors in the regulation of aldosterone production, we examined the effects of adenosine and adenosine agonists (N6-cyclohexyl adenosine; selective adenosine A1 receptor agonist and 5-N-ethylcarboxamine adenosine; selective adenosine A2 receptor agonist) on aldosterone and cyclic AMP production in rat adrenal capsular cells. Neither adenosine nor 5-N-ethylcarboxamine adenosine caused significant effects on basal aldosterone or cyclic AMP production. Also, adenosine (10–3M) showed no consistent effects on aldosterone and cyclic AMP production induced by ACTH. On the other hand, N6-cyclohexyl adenosine exhibited a significant inhibition of basal aldosterone and cyclic AMP production at doses of 10–4 M and 10–3 M; furthermore, 10–3 M N6-cyclohexyl adenosine inhibited aldosterone and cyclic AMP production stimulated by ACTH. These results suggest that adenosine A1 receptors are coupled to and inhibit adenylate cyclase and may be involved in the inhibition of aldosterone production.  相似文献

Ecdysteroids are hormones controlling cell proliferation, growth and the developmental cycles of insects and other invertebrates1. They are occasionally present in various unrelated plants for no apparent reason; no phytohormonal function has yet been identified. In certain cases, ecdysteroids are accumulated to high levels in leaves, roots or seeds. Some ecdysteroid-containing plants have been known as medicinal plants for centuries. One of them,Leuzea carthamoides Iljin (Asteraceae), growing in Central Asia, contains 0.4% ecdysteroid in dry roots and 2% in seeds. A pharmacological preparation from this plant, Ecdisten, is already available as a commercial preparation for its anabolic, tonic and other effects, for medical use (review2). It remained problematic, however, whether ecdysteroids were truly responsible for these effects, becauseLeuzea contains a number of other biologically active compounds in addition to ecdysteroids. We extracted and purified ecdysteroids from the seeds ofLeuzea. With 6 g of 96% 20-hydroxyecdysone (20E), we made a large-scale feeding assay with Japanese quail to find out whether ecdysteroid alone could duplicate the anabolic effects of the seeds. We found that the 96% ecdysteroid increased the mass of the developing quails in a dose-dependent manner, with the rate of increase proportional to the ecdysteroid content in the seeds; there was a 115% increase in living mass with 100 mg kg–1 of pure 20E compared with 109.5% increase with 100–180 mg kg–1 20E equivalents in the seeds. We conclude that the plethora of growth-promoting, vitamin-like effects induced in vertebrates byLeuzea is mediated by ecdysteroids.  相似文献

Incubation of molting glands from the crayfishProcambarus clarkii (Y-organ) and the silkwormBombyx mori (prothoracic gland) with 23,24-[2H4]-2-deoxyecdysone resulted in the production of deutero-ecdysone; this biotransformation was inhibited in the presence of xanthurenic acid. When the experiments were performed under an18O2 atmosphere, the18O atom was introduced into ecdysone, as confirmed by mass spectrometry. We therefore suggest that xanthurenic acid inhibits P-450-dependent hydroxylation of 2-deoxyecdysone. However, deutero-2-deoxyecdysone was not converted to 3-dehydroecdysone when using Y-organs in vitro, although it is a major product. We therefore conclude that the biosynthetic pathway of ecdysteroids inP. clarkii branches at an early step.  相似文献

Malaria, caused by members of the genusPlasmodia, is still the most prevalent parasitic disease in the world. In an attempt to understand genetic factors conferring resistance to malaria, mouse models of thalassemia, sickle trait, and ankyrin and spectrin deficiency were studied during infection with species of malaria infectious to rodents. Although growth ofP. falciparum is not inhibited in thalassemic erythrocytes in culture, mice carrying a -thalassemia mutation were protected fromPlasmodium chabaudi adami, supporting epidemiologic findings. Transgenic mice expressing s hemoglobin were also significantly protected from two species of rodent malaria. Importantly, a significant role for the spleen in protection in the s transgenic mice was found. Finally, mice deficient in spectrin and ankyrin were studied with respect to their ability to support the growth of malaria. It was found that spectrin deficient mice were almost completely refractory toP. chabaudi adami andP. berghei. These models will allow further study of host factors in resistance to malaria.  相似文献

To understand the role of calcium ions in thigmotaxis inParamecium caudatum, the effects of caffeine, ruthenium red and lanthanum (LaCl3) on thigmotaxis were examined. Thigmotaxis in the CNR mutant, which lacks voltage-dependent Ca2+-channels in the ciliary membrane, was also examined. Ruthenium red and LaCl3 suppressed thigmotaxis inP. caudatum, while caffeine enhanced it. The CNR mutant showed hardly any thigmotaxis. It can be thought that an increase in Ca2+ influx and the intraciliary concentration of Ca2+ ions induces thigmotaxis inParamecium.  相似文献

The proton-dependent synthesis of ATP was demonstrated in representative members of the generaHalobacterium, Haloarcula, andHaloferax. In all cases, synthesis was not inhibited by nitrate or N-ethylmaleimide, inhibitors of the vacuolar-like ATPase found in Archaea, but was affected by azide, an inhibitor of F0F1-ATP syntheses. These observations extend the earlier observations withHalobacterium saccharovorum and suggest that ATP synthesis in these organisms is brought about by an F0F1-APT synthase.  相似文献

The participation of protein kinase C (PKC) in the regeneration of tentacles ofHydra vulgaris was studied. Regeneration was induced by 1,2-sn-dioctanoyl-glycerol (diC8) and the novel diterpenoidic diacylglycerol verrucosin B (VB), a potent PKC activator extracted from marine sources. VB substantially increasedHydra average tentacle number (ATN) at concentrations 10,000 times lower than those needed for diC8 to exert an analogous effect. When both synthetic and natural VB analogues were tested, the structure/activity relationship found inHydra tentacle regeneration was identical to that known for DAG-induced activation of PKC in vitro. VB-induced increase of ATN was strongly counteracted by the PKC inhibitors sphingosine and A3, but was not synergic with a tenfold increase of extracellular Ca2+ concentration or with an increase of intracellular Ca2+ concentration obtained either with the ionophore A23187 or with thapsigargin. This suggested the involvement of a non-Ca2+-dependent PKC in VB-triggeredHydra tentacle regeneration. The involvement of phospholipase A2 (PLA2) activation inHydra regenerative processes was studied using the novel site-specific inhibitor of the enzyme, oleyloxyethylphosphorylcholine (OOPC), which brought about a striking inhibition of ATN in the low molar range. This effect was reversed by arachidonic acid (AA), while an enhancement of ATN was also observed with an inhibitor of AA uptake from membrane phospholipids, thus suggesting that PLA2-catalysed liberation of AA is involved inHydra tentacle regeneration. OOPC also blocked verrucosin B-induced PKC-mediated enhancement of ATN, thus suggesting that this effect is also mediated by PLA2 activation. ATN was increased also by compound 48/80, a direct activator of pertussis toxin-sensitive GTP-binding proteins, and this effect was counteracted by pertussis toxin pretreatment. None of the known AA cascade inhibitors exhibited an effect on ATN comparable to that exerted by OOPC, but, surprisingly, the cycloxygenase inhibitor indomethacin strongly enhanced ATN, thus suggesting that prostanoids might effect a negative control onHydra regenerative processes. This represents the first attempt so far reported to study the implication of more than one biochemical pathway as a signalling event in the hydroid regenerative processes.  相似文献

SelectedRhizobium bacteria, arbuscular mycorrhiza-forming (AM) fungi and associative bacteria have been shown to stimulate the growth of legumes, gramineae and cruciferae in field experiments on different soil types in temperate regions. A combination of microorganisms with different metabolic capacities (N2-fixation, P-mobilization; production of phytohormones and antibiotics) can partly surpass the effect of single inoculations, or can produce a positive effect where single inoculations are ineffective. Growth stimulation by inoculation requires microorganisms with phytoeffective metabolic characteristics and the ability to survive in the rhizosphere during the growth period. Another prerequisite is an adequate supply of plant assimilates for the production of microbial phytoeffective metabolites. Type of inoculum, method of inoculation and agricultural measures can influence the effect of the inoculation. Research is necessary to extend our knowledge both of basic principles, and about using microorganisms in practice.  相似文献

Adult morphology ofTrichogramma is modified by environmental conditions during the preimaginal development. For instance, the low temperatures inducing diapause cause a decrease of the ratio length of longest seta on male flagellum/width of flagellum inT. evanescens. The species of the host also influences numerous body ratios. So, in females, the development of tibiae with regard to wings is more important withScotia ipsilon than withGalleria mellonella. These results confirm the necessity of performing comparative morphological studies on individuals reared in the same conditions.  相似文献

Summary The-amylase gene ofDrosophila miranda is located on the X2-and on the neo-Y-chromosome, both developing sex chromosomes. Crosses between strains carrying different electrophoretically distinguishable alleles of the-amylase gene were performed. Females of the F1 offspring showed the expected heterozygosity, while the males proved to be hemizygous for this locus. Only the gene on the X2-chromosome is expressed, whereas the corresponding gene on the neo-Y-chromosome is not. Estimates of the-amylase activity in crude homogenates of male and female flies suggest strongly that the-amylase gene is dosage compensated inD. miranda. In contrast to this situation, in all otherDrosophila species the-amylase allele is autosomal and hence not dosage compensated.Acknowledgments. We would like to thank Betty C.
