The localized control of second messenger levels sculpts dynamic and persistent changes in cell physiology and structure. Inositol trisphosphate [Ins(1,4,5)P 3] 3-kinases (ITPKs) phosphorylate the intracellular second messenger Ins(1,4,5)P 3. These enzymes terminate the signal to release Ca2+ from the endoplasmic reticulum and produce the messenger inositol tetrakisphosphate [Ins(1,3,4,5)P 4]. Independent of their enzymatic activity, ITPKs regulate the microstructure of the actin cytoskeleton. The immune phenotypes of ITPK knockout mice raise new questions about how ITPKs control inositol phosphate lifetimes within spatial and temporal domains during lymphocyte maturation. The intense concentration of ITPK on actin inside the dendritic spines of pyramidal neurons suggests a role in signal integration and structural plasticity in the dendrite, and mice lacking neuronal ITPK exhibit memory deficits. Thus, the molecular and anatomical features of ITPKs allow them to regulate the spatiotemporal properties of intracellular signals, leading to the formation of persistent molecular memories.  相似文献

Alcohol dehydrogenase 3 (ADH3) has been assigned a role in nitric oxide homeostasis due to its function as an S-nitrosoglutathione reductase. As altered S-nitrosoglutathione levels are often associated with disease, compounds that modulate ADH3 activity might be of therapeutic interest. We performed a virtual screening with molecular dockings of more than 40,000 compounds into the active site of human ADH3. A novel knowledge-based scoring method was used to rank compounds, and several compounds that were not known to interact with ADH3 were tested in vitro. Two of these showed substrate activity (9-decen-1-ol and dodecyltetraglycol), where calculated binding scoring energies correlated well with the logarithm of the k cat/K m values for the substrates. Two compounds showed inhibition capacity (deoxycholic acid and doxorubicin), and with these data three different lines for specific inhibitors for ADH3 are suggested: fatty acids, glutathione analogs, and cholic acids.  相似文献

Summary Radiolabeled vitamin D3 was converted into several polar metabolites upon incubation with tissue homogenates from the common musselMytilus edulis. Chromatographic analysis indicated that the metabolites have chromatographic mobilities different from those of known standards. The results suggest that vitamin D3 is metabolized in mussels via pathways that differ from the vertebrate systems.Acknowledgments. The authors thank Dr Pirjo Rantamäki for the mussels and F. Genetic engineering, coupled with spectro scopic analyses, has enabled the metal binding proper ties of the α and β subunits of mouse metallothionein 1 (MT) to be characterized. A heterologous expression system in E.coli has led to high yields of their pure zinc-complexed forms. The cadmium(II) binding properties of recombinant Zn4-αMT and Zn3-βMT have been studied by electronic absorption and circular dichroism. The former binds Cd(II) identically to α fragments obtained from mammalian organs, showing that the recombinant polypeptide behaves like the na tive protein. Titration of Zn3-βMT with CdCl2 results in the formation of Cd3-βMT. The addition of excess Cd(II) leads to Cd4-βMT which, with the extra loading of Cd(II), unravels to give rise isodichroically to Cd9-βMT. The effect of cadmium-displaced Zn(II) ions and excess Cd(II) above the full metal occupancy of three has been studied using Chelex-100. Summary Plasma membrane sheets prepared by zonal centrifugation of a premicrosomal pellet obtained from a rat liver homogenate are devoid of HCO 3 -ATPase activity. Since the microsomal fraction is also lacking in this ATPase activity, it can be concluded that the HCO 3 -ATPase is not involved in the secretion of HCO 3 into bile. Acknowledgments. Summary The soil fumigant 1,2-dibromo-3-chloropropane (I) undergoes hydrolysis in the rat to a series of epoxide metabolites. Alkylation of glutathione by these epoxides produces 2 urinary metabolites identified as the mercapturic acids VI (R=COCH3) and VII (R=COCH3). Hydrolysis of the epoxides produces the male antifertility agentsa-chlorohydrin (IX, X=Cl) anda-bromohydrin (IX, X=Br) which are oxidatively metabolized to oxalic acid (XII), thus causing renal damage. These metabolic pathways can explain the toxic nature of the fumigant as a carcinogen, a male chemosterilant and as an agent causing kidney damage.This work was supported in part by the National Health and Medical Research Council of Australia.  相似文献

Summary 3-Dehydroretinal (vitamin A2 aldehyde) was found in the eyes of three species among 10 species of freshwater crayfish examined. Since dark-adapted eyes contained the 11-cis form of 3-dehydroretinal, this compound must be the chromophore of the visual pigment. 3-Dehydroretinal always coexisted with retinal (vitamin A1 aldehyde), indicating the presence of a rhodopsin-porphyropsin visual pigment system.  相似文献

The possible promoting effect of streptozotocin (STZ; 65 mg/kg body weight, intraperitoneal)-induced diabetes during 2-acetylaminofluorene (2-AAF; 0.04% in basal diet)-initiated hepatocarcinogenesis and modulatory effect of 1α,25-dihydroxyvitamin D3 (VD3; 0.3 μg/0.1 ml in propylene glycol, per os) were investigated by monitoring chromosomal aberrations (CAs), DNA strand breaks and specific DNA adducts in rat liver. VD3 treatment (twice a week) was started 4 weeks before the 2-AAF regimen and continued throughout the study. Aberrant metaphase chromosomes were counted from the regenerating hepatocytes 15, 30 or 45 weeks after STZ injection, while DNA strand break and adduct assays were performed 45 days post-STZ treatment. Dietary exposure to 2-AAF elicited a substantial increase in CAs and elevated the extent of DNA strand breaks and formation of N-(deoxyguanosin-8-yl)-2-aminofluorene. A promoting effect of STZ was evident from CAs coupled with DNA strand break analysis. VD3 treatment substantially reducted 2-AAF+STZ-induced CAs as well as DNA strand breaks and adducts. Thus, VD3 appears to be effective in suppressing liver-specific early chromosomal as well as DNA damage during the process of rat hepatocarcinogenesis initiated with 2-AAF and promoted by STZ contributing to its promise as a cancer chemotherapeutic agent. Résumé Nous avons étudié l'adsorption d'échange entre des précipités de BaSO4 et des solutions de BaCl2, Pb(NO3)2 et CaCl2 à l'aide de «tracers». Nous avons trouvé que l'adsorption peut être représentée par l'équation (1) pour l'adsorption d'échange; la capacité d'adsorption de notre BaSO4 était égale à 1,4 × 10–5 éq./g. Les grandeurs caractéristiquesf, figurant dans l'équation pour l'adsorption d'échange, avaient les valeursf(Ba)=1;f(Pb)=0,44;f(Ca)=0,00137.
