Elicitor prepared from the cell walls of Peni-cillium citrinum induces multiple responses of Taxuschinensis cells, including nitric oxide (NO) generation, se-quentially followed by the activation of PAL and synthesis oftaxol. NO scavenger cPITO and nitric oxide synthase (NOS)inhibitor PBITU prevent the latter two reactions, all of whichare triggered in the absence of elicitor by NO donor sodiumnitroprusside (SNP). The elicitor-induced NO release ofTaxus chinensis suspension cells is strongly inhibited byPBITU. These results demonstrate a causal relationship be-tween NO generation and the latter two reactions of Taxus chinensis cells to the elicitor, and also indicate that NO, pro-duced via NOS in Taxus chinensis cells treated with fungalelicitor, might act as an essential signaling molecule for trig-gering the activation of PAL and synthesis of taxol.  相似文献

The protein elicitor PB90 secreted by Phytophthora boehmeriae is an efficient elicitor inducing the hypersensitive response and systemic acquired resistance in tobacco plants. Here, we observed cell death in suspension-cultured cells of Nicotiana tabacum BY-2 with PB90 treatment using Trypan blue staining method. And this cell death could be suppressed by cycloheximide, an inhibitor of proteins synthesis, which implies that PB90-induced celldeath was an active cell death process requiring new protein synthesis. DAPI staining revealed that PB90 induce rapid chromatin condensation, margination, apoptotic bodies‘ formation and DNA laddering, further TUNEL assay also observed the specific breakage of 3 ‘-OH ends. All of the above common morphological characteristics indicated that PB90 induced apoptosis in suspension cultures of tobacco, suggesting that hypersensitive response induced by PB90 is an apoptotic process.  相似文献

The strategy of the two-component system,composed of Barnase and Barstar which encode RNase and a specific inhibitor to the RNase respectively, is adopted to obtain transgenic rice resistant to rice fungal blast disease. In this study, two chimeric promoters, induced by rice blast fungus pathogen (Magnaporthe grisea), are fused with Barnase respectively to construct two plant expression vectors, pWBNBS and pPBNBS together with the Barstar driven by CaMV 35S promoter. The resistance of the transgenic rice lines to rice blast fungus disease and rice blight disease are evaluated. The results show that (1) the expression of Barnase is induced in rice leaves when inoculated with the spores of Magnaporthe grisea; (2) the induced expression level of Barnase surpasses the level of Barstar, which elicits a similar hypersensitive response (HR) in the leaves, and the transgenic plant shows high resistance to the rice fungal blast disease; and (3) transgenic rice plants also show obvious resistance to rice bacterial blight disease. Taken together, these results suggest that the transgenic rice plants harboring this two-component system acquire relatively broad spectrum resistance against pathogens, especially high resistance to rice fungal pathogen.  相似文献

Enhancement of cell growth in suspension cultures is urgently needed in plant cell culture engineering. This study investigates the relationship between morphological transformation and cell growth in callus and suspension cultures of saffron cells belonging to the cell line C96 induced from Crocus safivus L In the suspension culture, an unbalanced osmotic pressure between the intracell and extracell regions induced a large morphological transformation which affected normal division of the saffron cells. An increase in osmotic pressure caused by the addition of sucrose inhibits the vacuolation and shrinkage of cytoplasm in the cells. As the sucrose concentration increases, the total amount of accumulated biomass also increases. Besides the sucrose concentration, increased ionic strength and inoculation ratio also help restrain to a large extent the vacuolation and shrinkage of the cytoplasm in the suspended cells, which results in increased biomass. The conditions for optimal biomass are: Murashige and Skoog's (MS) medium with 40 g/L sucrose and 60% (v/v) inoculation ratio.  相似文献

The effects of initial sucrose concentration, nitrate to ammonium ratio, total N concentration and phosphate concentration in medium on cell growth and isocamptothecin A and B synthesis by suspension call culture of Camptotheca acuminata were investigated in 250 mL shake flasks. 30 g L^-1 sucrose concentration was beneficial to secondary metabolites synthesis. The cell growth and metabolites synthesis were also affected by the ratio of NO3^-/NH4^+ , and nitrate was tavourable for cell growth. The maximum dry weight was achieved when nitrate was used as the sole N souree. The effect of total initial N on the cell cultures was also investigated with NO3^-/NH4^+ ratio of 1 : 2. The final dry cell weight was similar throughout culture period and 50 mM initial N was favourable for secondary metabolite synthesis. 50 mM initial phosphate concentration facilitated both cell growth and secondary metabolites synthesis.  相似文献

The spontaneous changes in cell shape and size, the membrane bending elastic modulus, and the structure and functions of intracellular hemoglobin of single human living erythrocyte incubated in different suspension pH were studied. The non-invasive, in situ, real time measurements on the parameters at the three levels of intracellular molecule, membrane and cell morphology were performed simultaneously by using a fast micro-image analyzing technology and a multi-channel micro-spectrophotometer system. The result shows that the cell shape and size, the mechanical property of the cell membrane, and the structure and function of intracellular hemoglobin are correlated and affected by each other with pH.  相似文献

The responses of rice to the second degree contamination of copper were studied by pot experiments under free-air CO2 enrichment (FACE) with 570 μmol ·mol^-1 of CO2. The results showed that the content of copper in rice leaves was reduced with the CO2 concentration reaching 570 μmol· mol^-1 and this happened more significantly under the second degree contamination of copper. Under FACE, activities of superoxide dismutase (SOD) enzyme in rice leaves treated by copper contamination were induced, whereas the contents of glutathione (GSH) and glutathione disulfide (GSSG) had no significant difference from the control. In the presence of ambient CO2, activities of SOD enzyme treated by copper pollution were suppressed during the whole rice growth, however, the contents of GSH and GSSG were induced at tillering and jointing stages, and then restored to the control levels in later growth under the second degree contamination of copper. With the rice growing, the content of malondialdehyde (MDA) rises continuously, but there had been no significant difference between the treatments at the same growth stage. Further studies are needed on the response mechanism of rice to Cu stress under elevated CO2.  相似文献

AFLP analysis of near-isogenic lines of the stripe rust resistance gene Yr10 was carried out with 6 PstⅠ- primers and 10 TaqⅠ-primers with the donor parent of Yr10 gene as the check. A total of about 4200 distinguishable bands were amplified, of which 5 were stable. The genetic linkage of the 5 polymorphic DNA fragments with the target gene were tested preliminarily on a segregating F2 population derived from a cross between the gene donor parent “Moro” and susceptible cultivar “Mingxian 169”. The DNA fragment PT0502 was found closely linked to the Yr10 gene and cloned and sequenced. Based on the sequence specific primers for PCR were designed and synthesized. Genetic linkage analysis with 195 segregating F2 plants indicated that the genetic distance was 0.5 cM between the main product SC200 fragment produced by PCR with the primers and the Yr10 gene. The primers can be used to detect the Yr10 gene quickly, effectively and exactly.  相似文献

Nitric oxide (NO) and Jasmonic acid (JA) are two key signaling molecules involved in many and diverse biological pathways in plants. Growing evidence suggested that NO signaling interacts with JA signaling. In this work, Our experiment showed that NO exists in guard cell of Vicia faba L., and NO is involved in signal transduction of JAinduced stomata closuring: ( i ) JA enhances NO synthesis in guard cell; ( ii ) both JA and NO induced stomatal closure, and had dose response to their effects; ( iU ) there are synergetic correlation between JA and lower NO concentration in regulation of stomatal movement; (iV) JA-induced stomatal closure was largely prevented by 2-phenyl-4,4,5,5-tetramethylimidazoline-l-oxyl-3-oxide (PTIO), a specific NO scavenger. An inhibitor of NO synthase (NOS) in mammalian cells, N^G-nitro-L-Arg-methyl eater (L-NAME) also inhibits plant NOS, repressing JA-induced NO generation and JA-induced stomatal closure. We presumed that NO mainly comes from NOS after JA treatment.  相似文献

The 660 bp region betweennodD3 P1 promoter and the following coding region ofRhizobium meliloti has been studied. This region is designated “downstream sequences”. It consists of two potential open reading frames, ORF1 and ORF2. Studies on the role of the downstream sequences on the activity ofnocD3 P1 with nod D3(P1)-IacZ fusion show that deletion of the sequences containing ORF2 causes the increase of the activity of the fusion; on the contrary, addition of extra copies of ORF2 markedly decreases the activity of the fusion. These results indicate that the product of ORF2 plays a negative role in the expression ofnod D3.  相似文献

The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a “whole genome shotgun” approach, we have produced a draft rice genome sequence ofOryza sativa ssp.indica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from anindica variant cultivar93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. The coverage of the draft sequence, the randomness of the sequence distribution, and the consistency of BIG-ASSEMBLER, a custom-designed software package used for the initial assembly, were verified rigorously by comparisons against finished BAC clone sequences from bothindica andjapanica strains, available from the public databases. Over all, 96.3% of full-length cDNAs, 96.4% of STS, STR, RFLP markers, 94.0% of ESTs and 94.9% unigene clusters were identified from the draft sequence. Our preliminary analysis on the data set shows that our rice draft sequence is consistent with the comman standard accepted by the genome sequencing community. The unconditional release of the draft to the public also undoubtedly provides a fundamental resource to the international scientific communities to facilitate genomic and genetic studies on rice biology.
