The Yunnan box turtle (Cuora yunnanensis, Boulenger, 1906), which has drawn much attention in conservation biology, was regarded as extinct since it was previously known only from 12 specimens collected in Yunnan, China, before 1908. Recently, live specimens have been discovered which are suggested to be C. yunnanensis. To determine whether the newly discovered specimens are really C. yunnanensis, we have established a molecular phylogeny, with a 1725-bp fragment of mitochondrial DNA, using samples from three live individuals of C. yunnanensis, together with sequence data from a museum specimen of C. yunnanensis (MNHN 1907.10) and other members of the genus Cuora. We found that the three newly discovered individuals and the old museum specimen of C. yunnanensis are very similar both in morphology and in mitochondrial DNA sequence, suggesting that the three new individuals are the very C. yunnanensis, and thus the species is not extinct. Our phylogenetic analysis also demonstrates that C. yunnanensis is not of recent hybrid origin, but rather represents a distinct evolutionary lineage.  相似文献

Based on a genetically modified radioresistant bacteria Deinococcus radiodurans, we constructed a real time whole cell biosensor to monitor radioactivity and genotoxicity in highly radioactive environment. The enhanced green fluorescence protein (eGFP) was fused to the promoter of the crucial DNA damage-inducible recA gene from D. radiodurans, and the consequent DNA fragment (PrecA-egfp) carried by plasmid was introduced into D. radiodurans R1 strain to obtain the biosensor strain DRG300. This engineered strain can express eGFP protein and generate fluorescence in induction of the recA gene promoter. Based on the correlation between fluorescence intensity and protein expression level in live D. radiodurans cells, we discovered that the fluorescence induction of strain DRG300 responds in a remarkable dose-dependent manner when treated with DNA damage sources such as gamma radiation and mitomycin C. It is encouraging to find the widely detective range and high sensitivity of this reconstructed strain comparing with other whole cell biosensors in former reports. These results suggest that the strain DRG300 is a potential whole cell biosensor to construct a detective system to monitor the biological hazards of radioactive and toxic pollutants in environment in real time.  相似文献

The short-lived reactive specimen nitrous acid HONO was generated in the gas phase by the heterogeneous reaction of gaseous HCl with AgNO2 which can generate higher concentration of HONO than other methods. We investigated the process from generation to dissociation in the gas phase under different controlled temperatures, and discussed the ionization and reaction on the solid surface by combination of the photoelectron spectroscopy and photoionization mass spectroscopy (PES-PIMS) and in situ diffuse reflectance infrared Fourier transform spectroscopy (DRIFTS).  相似文献

An exceptionally well-preserved fossil Kalligrammatid from the Jehol Biota   总被引:1,自引:0,他引:1  
A new species of Neuroptera, Sophogramma lii sp. nov. is described and illustrated in this paper. This new species is assigned to Sophogramma Ren of Kalligrammatidae, mainly based on its humeral recurrent veins, the basal cell present between posterior branch of media at base of wing, etc. The new species is distinguished from all other Sophogramma species on the difference of wing venation. This exceptionally well-preserved specimen is recovered from the Yixian Formation, Huangbanjigou, located near Beipiao City, in western Liaoning Province. The genus diagnosis has been emended based on new characters shown in this new species. Based on a summary of localities, distribution and geological ages of all genera and species of the Kalligrammatidae, we suggest that Kalligrammatids might have originated in Eastern Asia, especially in China. Function of the wing pigmentation is discussed briefly. The functional analysis of dr1127,a novel gene in Deinococcus radiodurans was performed in this pa-per. The dr1127 gene was found occasionally in our microarray and 2-DE gel experiments. Mutation of the dr1127 gene decreased the γ-radiation and H2O2 resistance of D. radiodurans,and weakened the scavenging abilities of cell extracts for free radicals (superoxide anion,hydrogen peroxide,and hy-droxyl radical). Further oxidative damage assays demonstrated that the purified DR1127 protein of D. radiodurans could bind to double stranded DNA in vitro and protect DNA from oxidative damage in this way. These results suggest that the dr1127 gene is an important gene that can maintain γ-radiation and oxidative resistance in D. radiodurans and may take part in the oxidative stress process.  相似文献

Biosynthesis and secretion of prothoracicotropic hormone (PTTH) of diapause- and nondiapausedestined individuals in Helicoverpa armigera were studied using whole-mount immunocytochemistry and enzyme-linked immunosorbent assay (ELISA). The immunocytochemistry revealed that PTTH is expressed in two pairs of lateral neurosecretory cells of the brain. The presence of immunoreactivity has not significant difference between the brains of the diapause- and nondiapause-destined 6th instar larvae. However, the obvious differences of expressional pattern from day 4 pupae were observed between the two types. PTTH titers in hemolymph from the 6th instar larvae to pharate adults were measured by the ELISA. Although there were similar titer changes between the two types of individuals at the larval stage, a significant difference from developmental expression was detected at the pupal stage, suggesting that the expression and secretion of PTTH does play a crucial role in regulation of pupal diapause of H. armigera.  相似文献

In eukaryotes, the Mre11-Rad50-Nbs1 (MRN) complex, which resides at the crossroads of DNA repair and checkpoint signaling, rapidly forms prominent foci at damage sites following double-strand break (DSB) induction. This complex carries out the initial processing of the DSB ends. Mutations in the genes that encode components of this complex result in DNA-damage hypersensitivity, genomic in- stability, telomere shortening, and aberrant meiosis. Therefore, the MR proteins are highly conserved during evolution. The bacterial orthologs of Mre11 and Rad50 are the SbcD and SbcC proteins, respec- tively. Deinococcus radiodurans, an extremely radioresistant bacterium, is able to mend hundreds of radiation-induced DSBs. The SbcD and SbcC proteins were identified as the products of the Dr1921 and Dr1922 genes. Disruption of the sbcD gene, by direct reverse-orientation insertional mutagenesis tech- nology, remarkably increases the cells’ sensitivity to various types of DNA damaging agents, such as ionizing radiation, ultraviolet irradiation, hydrogen peroxide, and mitomycin C. We also provide evidence that the drSbcD protein plays an important role in both growth and DNA repair in this organ- ism, especially in repair of DSBs generated after cellular exposure to 6000 Gy of IR. These results demonstrate that the drSbcD protein plays an important role in DSBs repair in D. radiodurans.  相似文献

A kind of temperature-sensitive nanotube array membrane was developed by modifying gold-nanotube array membranes with poly(N-isopropylacrylamide) (PNIPAm). The permeation ability of the mem-branes at different temperatures was investigated using sodium fluorescein and quantum dots as probes. The results showed that the pore diameter of nanotube was changed due to the reversible response of PNIPAm-modified membranes to temperature, and then the permeation ability of the mem-branes was changed. The permeation of fluorescence probes was slow and even almost blocked at 25℃ (below the lower critical solution temperature, LCST), since PNIPAm formed expanded structures and decreased the pore size. While at 40℃ (above the LCST), the permeation was increased, since PNIPAm became compact structures and the pore diameter was increased. Furthermore, the permeation ability of the temperature-sensitive nanotube array membranes could be adjusted reversibly and it is possible to use the membranes in nanofluidic devices, nanogates, etc.  相似文献

Pheromone gland extracts of the fall webworm, Hyphantria cunea (Lepidoptera: Arctiidae) were analyzed by gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS). Based on comparison of retention times and mass spectra of synthetic standards, four compounds were identified as (9Z,12Z)-octadecadienal, (9Z,12Z,15Z)-octadecatrienal, (3Z,6Z,9S, 10R)-9,10-epoxy-3,6-heneico- sadiene and (3Z,6Z,9S,10R)-9,10-epoxy-1,3,6-heneicosatriene with a ratio of 2:33.6:58.4:6. Trapping tests showed that quaternary blend of these compounds was highly attractive to male H. cunea. Unitraps baited with the quaternary blend loading of 200-300 μg on a gray rubber septum were very effective for monitoring H. cunea populations.  相似文献

The rate constants for the ozone reactions with n-butyl methyl sulfide (n-BMS, CHaCH2CH2CH2SCH3), sec-butyl methyl sulfide (s-BMS, CH3CH2(CH3)CHSCHa) and tert-butyl methyl sulfide (t-BMS, (CH3)3CSCH3) were measured using our smog chamber under supposedly pseudo-first-order conditions at 30002 K and 760 Torr. The experimental determined rate constants for n-butyl, s-butyl and t-butyl methyl sulfide are (1.23 ± 0.06)×10-19, (5.08 ± 0.19)×10-20 and (2.26 ± 0.14)×10-20 cm3 molecule-1· s-1, respectively. The reactivity-structure relationship of the reactions was discussed and used to illustrate the mechanism of the ozone reaction with thioethers. The results enrich the kinetics data of atmospheric chemistry.  相似文献

Nano-SiC doped MgB2 tapes were prepared by the in situ powder-in-tube method. Heat treatment was performed at 650℃ for 1 h. XRD data indicate that SiC particles had reacted with the MgB2 during sintering process. MgB2 core seemed to be denser after SiC doping, and the critical temperature was slightly depressed. The critical current density Jc of the SiC doped tapes was significantly enhanced in magnetic fields up to 14 T compared to the undoped ones. For the 5% SiC doped samples, Jc was in- creased by a factor of 32 at 4.2 K, 10 T. The enhancement of Jc-B properties in SiC doped MgB2 tapes is considered to be due to the enhancement of grain linkages and the introduction of effective flux pining centers. The substitution of B by C in MgB2 grains is thought to be the main reason for the improve- ment of the flux pinning ability in SiC doped MgB2 tapes.  相似文献

Bionic asymmetry: from amiiform fish to undulating robotic fins   总被引:2,自引:0,他引:2  
Similar to bionic non-smooth which has been successfully applied in anti-resistance and anti-adhesion, bionic asymmetry is also an inherent property of biological systems and is worth exploring for conceivable pragmatic applications. Therefore, bionic asymmetry for undulations is of main interest in this paper. We initially investigate bionic asymmetry with a case study of the undulating robotic fin, RoboGnilos, which evolved from the long dorsal fin of Gymnarchus niloticus in the amiiform mode. Since the performance of the pre-existing undulating fins is hardly satisfactory, we obtain bionic inspirations of undulatory asymmetry through observations and measurements on the specimen of G. niloticus, to improve upon the performance. Consequently, the newly acquired innovation for bionic asymmetry is incorporated into the previously derived kinematics model, and also applied to the experimental prototype. Both computational and experimental results verify that bionic asymmetric undulation generates better propulsion performance (in terms of linear velocity and efficiency) than the traditional symmetric modes with the same undulatory parameters. The Na^+/H^+ antiporter plays key roles in maintaining low cytoplasmic NaNa^+ level and pH homeostasis, while little is known about the Carboxyl-terminal hydrophilic tails of prokaryotic antiporters. In our previous study, the first Na^+/H^+ antiporter gene nhaH from moderate halophiles was cloned from Halobacillus dabanensis D-8 by functional complementation. A topological model suggested that only nine amino acid residues (^395PLIKKLGMI403) existed in the hydrophilic C-terminal domain of NhaH. The C-terminal truncated mutant of NhaH was constructed by PCR strategy and designated as nhaH△C. Salt tolerance experiment demonstrated that the deletion of hydrophilic C-terminal nine amino acid residues significantly inhibited the complementation ability of E. coil KNabc, in which three main Na^+/H^+ antiporters nhaA, nhaB and chaA were deleted. Everted membrane vesicles prepared from E. coil KNabc/nhaHAC decreased both Na^+/H^+ and Li^+/H^+ exchange activities of NhaH, and also resulted in an acidic shift of its pH profile for Na^+, indicating a critical role of the short C-terminal domain of NhaH antiporter in alkali cation binding/translocation and pH sensing.  相似文献

A chiral complex of (R,R)-Pd was prepared by reaction of optically pure (R,R)-bis(pyrrol-2-ylmethyl-eneamino) cyclohexane ligand with Pd(OAc)2·2H2O under the base condition at room temperature. Countless fossil embryo Olivooides and the hatched larvae, juveniles and adults (the latter two kinds are Punctatus) are recovered by means of acid maceration from the fine-crystalline to medium-crystalline phosphatic limestone and phosphatic micrite of Early Cambrian Kuanchuanpu Formation at the Shizhonggou section, near Kuanchuanpu Village, Ningqiang County, Shaanxi Province, China. Using the technique of Synchrotron X-ray Tomographic Microscopy, the 3D internal structure of Ofivooides and Punctatus is reconstructed. The morphological and statistic analyses are also given to the stellae structure of Ofivooides and Punctatus, which indicates that this structure is a result of adaptive evolution to a lifestyle of fast-attaching after hatching, probably with the function of mucilage secretion. The internal structure of Punctatus is described and discussed. The ovum-like structure, a common internal feature of Punctatus, is considered as the taphonomic structure, rather than eggs or other biological structure. This structure is thought to be formed after the burial of the animal and before or during the mineralization. The original internal structure of Punctatus is assumed to be tabulae-filled, with soft body grown on them.  相似文献

Surface soil samples collected over a high spatial resolution in eastern China were analyzed for carbon isotope composition (δ^13C) of total organic carbon (TOC) and higher plant-derived long-chain n-alkanes, with the latter reported as weighted mean values. The two sets of δ^13C values are significantly correlated and show similar trends in spatial variation. The spatial distribution of δ^13C shows less negative values in the mid-latitudes between 31°N and 40°N and more negative ones at higher and lower latitudes. This is consistent with previously reported carbon isotope data from surface soil phytoliths in the same region and suggests that the mid-latitude area provides relatively favorable growing condi- tions for C4 plants. Furthermore, δ^13C values of both TOC and long-chain n-alkanes from 12 surface soil samples collected from a small grassland in north China displayed similar carbon isotope values and the difference between paired δ^13C of a soil samples remains relatively constant. Our data demonstrate that in eastern China, soil δ^13C composition of both TOC and long-chain n-alkanes is effective indicators of C3/C4 ratios of the prevailing vegetation. This work suggests that -22‰ and -32‰ are good es- timated end members for the weighted mean δ^13C values of long-chain n-alkanes (C27, C29 and C31 n-alkanes) from soils under dominant C4 or C3 vegetation, allowing us to reconstruct paleovegetation trends.  相似文献

The way of introducing sodium azide (NaN_3) into the reaction solution played an important role in the preparation of cBN by hydrothermal synthesis method. The results showed that both cBN content and crystalline perfection of the samples improved with increasing R_N value, and pure cBN could be obtained at 300℃ and 10 MPa when R_N increased to 3:1. Here R_N is defined as R_N =NaN_3(I)/NaN_3(II), wherNaN_3(I) denotes the amount of NaN_3 (in molar) that is added into the autoclave at the beginning of threaction process, and NaN_3(II) is the amount of NaN_3 (also in molar) introduced into the autoclave ahigh temperature and high pressure (i.e. 300℃ and 10 MPa). In order to explain the experimental results, a preliminary model was proposed in this paper.  相似文献
