Immunoreactive neurons and fibers were localized in the Rat brain by immunocytochemistry using an anti-ACTH 17-39 antiserum. Neither adrenalectomy nor water deprivation affected intensity of distribution of reaction of labelled neurons whereas colchicine increased labelling of hypothalamic perikarya. These results support the neuronal origin of cerebral ACTH.  相似文献

Retrograde axonal transport of Horseradish peroxydase (HRP) has been used to trace the cells of origin of proprioceptive fibers in jaw-closing and jaw-opening muscles. After injection of HRP in young cats' masticatory muscles (masseteric, temporal, pterygoid, mylohyoid and digastric) labelled neurons were found in the ipsilateral semi-lunar ganglion and trigeminal mesencephalic nucleus. It is concluded that sensory endings are present in jaw-opening as in jaw-closing muscles; possibly the afferent fibers from muscle endings of the opener muscles have their somata in the mesencephalic nucleus, afferent fibers from tendinous receptors in the semi-lunar ganglion.  相似文献

(1) Isolated livers of previously fasted fafa and Wistar Rats are perfused with labelled oleic acid and glycerol. Two pools of synthesized lipids are studied: --a pool of liver lipids: labelled TG 16:0, 16:0, 18:1, 16:0, 18:1, 18:1 and 18:1, 18:1, 18:1 are synthesized more by the fafa than by the Wistar Rat liver, The TG 16:0, 18:1, 18:2 appears in the liver of the 2 rat strains. After a 30 min. perfusion, the PL are synthesized less by the fafa than by Wistar Rat liver; --a pool of lipids released in the circulation. The labelled tG 16:0, 18:1, 18:2, are increased in the perfusate of fafa rat. The labelled TG 18:1, 18:1, 18:2 and 18:1, 18:2, 18:2 are in the same amount. (2) The liver synthesis of TG 12:0, 12:0, 12:0 is demonstrated in the fafa Rat by a lauric acid perfusion. This TG is not present in circulation, when a Wistar Rat liver synthesizes and releases with this TG, TG containing 1 or 2 molecules of lauric acid.  相似文献

After thymidine-3H injection into pregnant Rat, histoautoradiographic studies showed that adipose cell proliferation in different subcutaneous and visceral sites, was very important in Rat foetuses about 24 h before birth and that lipid accumulation in the labelled cells was very fast during perinatal 48 h.  相似文献

A fluorescence histochemical technique using glyoxylic acid and cryostat sections has been worked out to visualize the dopamine-containing neurons of the retina. It allows the demonstration of catecholaminergic junctional cells and alloganglion cells, classically described. Moreover, the observation of some fluorescent fibers in the external zone of the inner nuclear layer, in contact with the outer plexiform, suggests the presence of catecholamine-containing interplexiform cells, in the Rat retina.  相似文献

Lauric acid, labelled oleic acid and glycerol are perfused in isolated liver of fafa Rats and Wistar Rats previously subjected to fasting. They synthesize TG and PL de novo, though in long time experiments with the normal Rat, the most important method of synthesis is an exchange of AG of the endogenous glycerolipids. However PL are not synthesized with lauric acid. In the livers of fafa Rats the synthesis of TG with oleic acid and glycerol is higher than in livers of Wistar Rats: 16:0 18 : 1 18: 1, 16:0 18: 1 18: 2, 18 : 1 18 :1 18:1, 16 : 0 16 :0 18 : 1 (this TG is not present in liver of Wistar Rat). The hepatic synthesis of PL by the fafa Rat, is less important after 15 min while it is important with Wistar Rats. The synthesized TG with lauric acid (only the TG 12 : 0 12 : 0 12 : 0 with the fafa Rat) are more rapidly oxidized by liver of obese Rat than by liver of normal Rat.  相似文献

Summary Binucleated neurons were studied in the central nervous system of the rat and the rabbit. They were present in the young as well as the adult animals. The animals injected with thymidine-H3 during their embryonic development showed labelled binucleated neurons. It is suggested that the neurons become binucleated during neuroembryogenesis, and differentiate into normal neurons morphologically and physiologically.Supported by NIH Research grant No. NS-08817-05.  相似文献

The behaviour of cariogenic Bacteria (Streptococcus mutans) is studied with regard to collagen, which represents 90% of the dentine organic matrix. Collagenase activity of cariogenic Bacteria is measured with radioactive precursors and gel electrophoresis and compared to reference bacterial collagenase (Clostridium histolyticum). Labelled collagen substrate has been prepared with two different methods: extraction by 0,5 M acetic acid from young Rat skin, previously labelled with L-proline 14C, or reduction by Na B3H4. Both collagen sutstrates have been incubated for 2 h in Terleckyj medium in which the Streptococcus mutans have been inoculated. The experiments show a proteolytic activity of Streptococcus Mutans on the collagen substrate.  相似文献

The existence of autapses, i. e. of synapses between a neuron and its own axon, is demonstrated in vitro by means of Horseradish peroxidase (HRP) iontophoretic filling, combined with electron-microscopic (EM) analysis. After the injection of one single Purkinje cell per explant (Kitten cerebellum), the ultrastructural observation (serial thin sections) reveals the presence of both pre- and post-synaptic labelled structures. Axodendritic autapses are thus illustrated and their significance is discussed in view of earlier morphological and electrophysiological data.  相似文献

In the 10 day old hemicastrated Rat daily injections of testosterone (total doses of 0.04 to 4 mg/Rat) or of proteinaceous extract of Rat testis (4 mg/Rat) inhibit compensatory hypertrophy of the remaining testis. Both testosterone and testicular proteinaceous extract could be involved in the growing processes of the testis, locally or by means of modification of gonadotropin secretion. Results have been analysed by covariance analysis to eliminate the influence of body weight variations.  相似文献

The localisation of brain glycoprotein NSA 3 was studied by means of indirect immunofluorescence on alcohol fixed, paraffin embedded sections of Rat brain. These techniques allowed the localisation of NSA 3 to the membrane of some (about 10%) of the neurons. In the white matter, the patterns were in agreement with the localisation of the Ranvier nodes. The nodes of Ranvier were also stained in peripheral nerves.  相似文献

Microiontophoretic applications of DL-2-amino-3-phosphonopropionic, DL-3-amino-4-phosphonobutyric, aminomethylphosphonic, 2-aminoethylphosphonic and 3-aminopropylphosphonic acids in the course of extracellular recordings of central neurons in the Rat resulted in effects which appeared similar to those observed with their carboxylic analogs. Summary Rat offspring were maternally subjected to methadone hydrochloride during gestation or lactation. At 21 days of age, the area of the pyramis (cerebellar lobule VIII) in prenatally and postnatally exposed groups was reduced 45% and 36%, respectively, from control levels, and the total number of internal granule neurons/section was reduced 49% and 46%, respectively; the number of granule neurons/mm2 in both groups was 75% of control values. Based on histological evaluation the timetable of cerebellar morphogenesis was unaltered in rats prenatally exposed to methadone, suggesting a permanent neuronal deficit, but cerebellar development was markedly delayed in animals subjected postnatally.This work was supported by National Institute on Drug Abuse grant DA-01618.  相似文献

I and i antigens were detected by indirect immunofluorescence on human erythrocytes. 80% of the cord blood cells were brightly fluorescent with anti-i, 15% only with anti-I. In adult blood, this ratio is reversed, 2% only of the cells being intensively labelled with anti-i. The brain structures specifically involved in harmaline-induced tremor have been identified in the Rat by using the 14C-2-deoxyglucose marking Method. The results obtained in an animal treated with tremogenic doses of harmaline, but immobilized with Faxedil, have been compared with those of another animal, not treated, but submitted to the same experimental procedure. The most specifically marked structure was the inferior olive. Secondarily marked structures were the posterior part of the lateral reticular formation, the caudato-putamen area and the prefrontal and frontal cortex.  相似文献

Day 25 after insemination is a date of peculiar importance in the maturation of several organs in the Rabbit fetus. From day 25 onward the fetal liver stores increasing amounts of glycogen and the lung stores increasing amounts of lecithins, concomitant with sudden rise in the activity of lung phosphatidic-acid phosphohydrolase. Earlier studies on decapitated fetuses established that glycogen storage in the liver is dependent on a dual hormonal control, comprising a pituitary hormone like growth hormone or prolactin (some placental hormones share the same activity) and corticosteroids (Jost, 1961). Since the variations in endogenous corticosteroids do not seem to herald these liver or lung changes (Mulay et al., 1973), a study was made of growth hormone. Plasma immunoreactive growth hormone--determined with a heterologous Rat system (Kervran et al., 1976)--increases eightfold between days 23 and 25. During the same time plasma prolactin does not change according to McNeily and Friesen, 1978, and to unpublished data obtained with Dr McNeilly. In preliminary assays, Rat growth hormone was seen to increase phosphorylase "a" activity in the lung of 18.5 day-old Rat fetuses, thus anticipating normal development. We suggest that growth hormone plays a role in initiating liver and lung maturation.  相似文献

A simple method for counting nuclei in the preimplantation mouse embryo   总被引:4,自引:0,他引:4  
An easy and rapid method of counting the number of cells in the preimplantation mouse embryo is described. The procedure increases the speed with which large numbers of embryos can be processed using a simple squash technique. Cell numbers are determined by exposing the embryos to the fluorescent DNA-binding dye, Hoechst 33258, removing the zona pellucida and simply squashing the embryo and counting the number of fluorescent nuclei. An increase in fluorescent intensity and maintenance of nuclear conformation of the squashed preparations are greatly improved by the use of the non-ionic detergent Triton X-100. Viability of dye-treated fertilized one-cell and blastocyst stage embryos is maintained at least up to day 13 of pregnancy following transfer of the embryos to the uteri of pseudopregnant recipients. Additional uses for this staining technique are discussed.  相似文献
