Expression of alpha-cardiac myosin heavy chain in mammalian skeletal muscle.

We have investigated the reactivity of different human, rat and cat muscles to a monoclonal antibody directed against human alpha-cardiac myosin heavy chain. We have found that special fiber subpopulations of human masseter and extraocular muscles, as well as the bag fibers of human, rat and cat muscle spindles, were reactive to this antibody, indicating that these fibers expressed alpha-cardiac myosin heavy chain or a closely related isoform. This isomyosin was present in the spindle bag fibers at early fetal stages, whereas its expression in masseter and extraocular muscle fibers was not detected during the first 22 weeks of gestation. Our results add to the list of muscle proteins which are expressed in locations or at developmental stages other than those initially described, suggesting that a revision of the present nomenclature of the subgroups of myosin heavy chains might be considered in the future.     

Exchange of tritium from randomly tritiated taurocholate by microbial bile salt oxidoreductases

Summary Tritium distribution on randomly labelled taurocholate (TC) was estimated at 28%, 4%, 1% and <0.5% on the hydrogens opposite the 3-, 7- and 12-OH groups and taurine moiety respectively. Anomalously,C. perfringens 3-hydroxysteroid dehydrogenase (3-HSDH) catalyzed tritium loss of 36% on formation of 7-, 12-dihydroxy-3-keto-5-cholanoate, implying additional losses of tritium at other sites by this enzyme.This work is supported by the National Cancer Institute.     

Incorporation of α-aminobutyric acid into ergostine byClaviceps purpurea

Summary Claviceps purpurea 275 F.I. fed with -aminobutyric acid produces ergostine with the corresponding -hydroxy--aminoacid undiluted by endogenous aminoacid. Almost all the -aminobutyric acid not incorporated into the ergostine remains unchanged in the medium. The significance of these results in the biosynthesis of ergot alkaloids is discussed.     

Age- and sex-related differences in the content of prothymosinα in rat tissues

Differences in the tissue content of prothymosin during the early postnatal development of male and female rats are reported. Thymus and spleen have been found to contain significantly higher amounts of prothymosin in the newborn and prepubertal animals, as compared to adults, whereas liver has been found to contain low levels of prothymosin throughout development. These findings indicate a functional association of prothymosin with the proliferating lymphoid tissues of the young rat.     

Interdependence between locomotor activity and duration of wakefulness in humans during isolation

Summary Locomotor activity was recorded in 14 subjects who lived singly in an isolation unit for 16 to 88 days. Their free-running circadian rhythms had a mean period of 25.9 h, with individual means in the duration of wakefulness () ranging from 12.1 to 22.9 h. Intraindividually, the hourly means of activity were negatively correlated with to such a degree that the total amount of activity per day remained constant irrespective of large variations in .

17β-Hydroxymethyl-Steroide: Darstellung von Derivaten des 17α-Hydroxy-17β-hydroxymethyl-4-androsten-3-ons

Summary Starting from 17-hydroxy-17-hydroxymethyl-4-androsten-3-one, we synthesized 4-chloro-17-hydroxy-17-hydroxymethyl-4-androsten-3-one-17, 20-acetonide and 17-hydroxy-17-hydroxymethyl-1, 4-androstadien-3-one. 11, 17-dihydroxy-17-hydroxymethyl-4-androsten-3-one was obtained from cortisone via methyl-17-hydroxy-3, 11-dioxo-4-etienate.     

The synthesis of corticosteroids methylated at C21

Zusammenfassung Die Darstellung der beiden epimeren C₂₁-Methylderivate des 9-Fluorprednisolons wird beschrieben. 9-Fluor-21-methyl-1, 4-pregnadien-11, 17, 21B-triol-3, 20-dion-21-azetat ist ein hochwirksames entzündungshemmendes Steroid, vollkommen frei von Nebenwirkungen der Natrium- und Wasserretention des C₂₁-unsubstituierten 9-Fluorprednisolons.     

Levels ofα 1 in the spring and autumn seasons

Summary In a group of 84 pairs of 11-year-old children of both sexes, the level of the ₁-antitrypsin ( ₁-AT) were ascertained in the autumn and spring. Although the mean levels of ₁-AT in the two seasons hardly differed, the highly significant seasonal changes in the distribution curves of ₁-AT values were noted in boys, whereas the levels showed higher stability in girls.

Summary In vitro, the mixture Chlorpromazine-human serum gives modifications of the lipoproteins and the-globulins. These modifications are studied by paper electrophoresis, Tiselius-electrophoresis, and immuno-electrophoresis. Under the experimental conditions described, the selective precipitation of the ₁-globulins seems quite unexpected.     

Distribution of C19-steroid conjugates in plasma protein fractions

Zusammenfassung Nach. i.v. Gabe von 7-³H-Dehydroepiandrosteron wurde menschliches Plasma einer präparativen Zonenelektrophorese unterworfen und in Albumine, ₁-, ₂-,- und-Globuline zerlegt. Aus den einzelnen Proteinfraktionen trennte man freie Steroide, Steroid-Sulfokonjugate und -Glukuronoside ab und untersuchte letztere Fraktionen auf ihren Gehalt an markierten C₁₉-Steroiden. Es zeigte sich, dass die Steroid-Sulfokonjugate vornehmlich in der Proteinfraktion der Albumine, die Steroid-Glukuronoside dagegen bevorzugt in der Proteinfraktion der ₁-Globuline auftraten.     

Fehlende Wasserstoff-Übertragung von Östradiol auf Androstendion im Diäthylnitrosamin-induzierten Hepatom der Ratte

Summary In liver-tissue of female rats estradiol-17 -T is oxydized by NAD or NADP, yielding the tritiumlabelled coenzymes NAD-T or NADP-T which can transfer the tritium to androstendione. So tritium-labelled androstandiol is obtained. Of the added activity 10–30% is oxydized to water. In diethylnitrosamine-induced hepatomas estradiol-17 -T is oxydized to tritium-water in a comparatively small amount and no transport of the tritium to reduction-products of androstendions occurs. Testing the in-vivo-oxydation of estradiol in normal female rats and in females with hepatoma with the aid of the Stoffwechsel-labil labelling (measuring the specific activity of tritium in body-water after injecting estradiol-17 -T) a lower oxydation-rate was also found in rats with hepatoma than in normal rats.     

The tumor necrosis factor α of the bony fish seabream exhibits the in vivo proinflammatory and proliferative activities of its mammalian counterparts,yet it functions in a species-specific manner

Information on the bioactivities of non-mammalian cytokines is scant due to the lack of the recombinant molecules and specific antibodies. We produced the mature predicted peptide of tumor necrosis factor (TNF) from the bony fish gilthead seabream (Sparus aurata L.) (sbTNF), and its biological role was determined in vitro and in vivo. We first demonstrated by analytical size-exclusion chromatography that sbTNF is an oligomeric protein but the dimer appears to predominate over the trimeric form, in contrast to mammalian TNF. Intraperitoneal injection of native sbTNF resulted in (i) priming of the respiratory burst of the peritoneal exudate and head-kidney (HK) leukocytes, the latter being the bone marrow equivalent in fish; (ii) rapid recruitment of phagocytic granulocytes to the injection site, and (iii) induction of granulopoiesis in the HK. Interestingly, sbTNF was able to induce a strong proliferation of HK cells in vitro, whereas human TNF did not. Conversely, sbTNF was not cytotoxic for murine L929 fibroblasts.Received 12 February 2004; received after revision 15 March 2004; accepted 29 March 2004

Summary The oestrogenic activity of -bromo-,-diphenyl--pentadeuteriophenylethylene is compared with that of the non-deuterated analogue, and found to be significantly lower. No sizeable difference was found between the two compounds as regards their preoestrus-inducing activity.

Defensive steroids from a carrion beetle (Silpha americana)

Summary The defensive anal effluent discharged bySilpha americana in response to disturbance contains a mixture of steroids stemming from a glandular annex of the rectum. The compounds have been characterized as 15-hydroxyprogesterone (1, principal component), 5-pregnan-15-ol-3, 20-dione (2), 5-pregnan-3, 15-diol-20-one (3), 5-pregnan-7, 15-diol-3,20-dione (4), 5-pregnan-3, 7, 15-triol-20-one (5), 5-pregnan-16-ol-3,20-dione (6), and 5-pregnan-3, 16-diol-20-one (7), none previously found in insects. Bioassays with jumping spiders showed compounds1 and6 to be feeding deterrents at the 1 g level.Paper No. 78 of the series: Defense Mechanisms of Arthropods. Positive correlation of responsiveness to catecholamines of the rat liver glycogenolytic receptor with other α-receptor responses

Zusammenfassung Durch die vollständige Dosis-Wirkungskurve der Katecholamin-i.p.-induzierten Leber-Glykogenolyse an der Ratte konnte deren Einfluss auf die -Rezeptoren nachgewiesen werden. Da eine Wirkung auf die -Rezeptoren (Lipolyse) ausschied, konnte es sich nur um einen -Effekt handeln.     

Hydrolysis of N-phenylacetyl-α-methyl-α-amino acids by benzylpenicillinacylase

Summary Enzymatic hydrolysis of several racemic N-phenylacetyl--methyl--amino acids containing an additinal aliphatic, aromatic or polar substitutent on the chiral carbon atom, has been studied by using benzylpenicillincylase fromEscherichia coli A.T.C.C. 9637. Both the rate of hydrolysis and the stereoselectivity were found to be considerably lower than in the case of natural -amino acids. Steric and electronic factors in the side chains influencing the stereoselectivity are discussed.
