Summary Chronic administration of potassium canrenoate (SC-14266), a water-soluble anti-aldosterone agent, increased serum LH levels in male rats without altering serum levels of FSH, prolactin, testosterone and progesterone. Summary Pituitary levels of prolactin were significantly diminished after 5 s.c. injections spaced 12 h apart of 5 g of a partially purified bovine pineal peptide fraction (E5) in both intact and castrated rats. E5 treatment did not significantly affect the castration-induced changes in plasma luteinizing hormone (LH), follicle-stimulating hormone (FSH) or pituitary LH but did partially block the pituitary fall in FSH in castrated animals.Supported by NSF grant PCM 8003441 and NIH Center for Reproductive Biology (Bioassay Core) No.P30 HD 10202. Summary Treatment of orchidectomized rats with LH, FSH or prolactin decreased the tubulin content of the medial basal hypothalamus (MBH), whereas FSH or prolactin augmented it in the adenohypophysis (AH). After castration, negative correlations existed between serum LH and total or polymerized MBH tubulin, whereas in the AH positive correlations were found. Serum levels of testosterone, FSH, LH and prolactin were measured by RIA in patients with BPH (benign prostatic hypertrophy) and in control men. The testosterone concentrations in the BPH group were significantly lower than that of the control group. The aim of this study was to compare the effects of pineal indole treatments on LH and FSH release in pinealectomized and suprachiasmatic lesioned and ovariectomized rats rendered hyperprolactinemic by acute sulpiride treatment. pinealectomy or suprachiasmatic nuclei lesions in female rats both decreased plasma LH and FHS at 10, but not at 20 d after surgery, whereas the daily afternoon administration of melatonin effectively restored levels of both gonadotropins to control values. In ovariectomized rats, pinealectomy or suprachiasmatic nuclei lesions were ineffective in counteracting the high plasma levels of LH and FSH. However, sulpiride treatment in both pinealectomized and suprachiasmatic nuclei lesioned and castrated female rats significantly decreased the levels of LH and FSH, an effect which was counteracted by daily afternoon melatonin administration. Other pineal indoles tested, i.e., 5-hydroxy- and 5-methoxytryptophol, were ineffective in regulating gonadotropin levels. The results suggest that the pineal gland, through its hormone melatonin, can modulate gonadotropin secretion by acting on a dopamine mechanism independent of hypothalamic suprachiasmatic areas.  相似文献

Summary In prolactin (PRL)-deficient male dwarf mice, treatment with PRL stimulates the release of FSH without affecting plasma LH levels. We now report that this effect of PRL is not mediated by the testes and that PRL does not modify FSH or LH release in female dwarf mice.This work was supported by the National Institute of Child Health and Human Development through a grand HD12642 and RIA Core of grant HD10202. The effect of pinealectomy on the circadian periodicity in serum gonadotropins was investigated in adult male rats. Pinealectomy resulted in an elevation of the serum FSH concentration 7 days after the operation. A 4-fold increase in serum FSH over 24 h with no concommitant increase in serum LH following pinealectomy suggests that the control of FSH secretion could be mediated via the pineal gland.  相似文献

Resumen La administración continuada de-metilpara-tirosina, un inhibidor de la síntesis de catecolaminas, impide la hipertrofia compensadora del ovario en ratas. Este efecto es prevenido por la simultânea injección de DOPA, un. precursor de las catecolaminas. El grado de la hipertrofia compensadora del ovario no fue modificado por lap-cloro-fenilalanina, un inhibidor de la síntesis de serotonina. El estudio histológico de los ovarios de los animales tratados con MPT sugiere que ésta bloquea la ovulación, no modificando aparentemente la secreción tonica de las gonadotrofinas FSH y LH.

Summary A new piperidine derivative, the 2-diphenylmethylpiperidine, produces already in doses of 0.001 g/kg s.c. a strong increase of the spontaneous motility of various animals. On mice it has about the same potency as d-desoxy-ephedrine and is about 3 times more potent than amphetamin. It also acts markedly antagonistic on various narcotics but has practically no peripheral sympathomimetic effects.

Summary Plasma concentrations of gonadotropin, prolactin and hypothalamic tyrosine hydroxylase (TH) activity were measured in ovariectomized rats treated with aminooxyacetic acid (AOAA), a drug which elevates brain GABA levels. Hypothalamic TH activity was significantly increased with a significant decrease in prolactin (Prl) release. Plasma levels of gonadotropins were not modified by AOAA. These results support an inhibitory action of GABA on Prl release possibly mediated through hypothalamic dopamine.Supported by grants from Indian Council of Medical Research (ICMR), New Delhi. Previous studies indicate that the pineal gland alters prolactin secretion, and it was suggested that at least part of the effect of the pineal hormone melatonin on prolactin release may be mediated by the hypothalamic structures. In this study, pinealectomy and lesions of the suprachiasmatic nuclei were found to alter serum levels of prolactin in the same direction, an effect that was counteracted by daily afternoon melatonin administration. Melatonin, but not other pineal indoles, also prevented sulpiride-induced prolactin secretion in pinealectomized or suprachiasmatic nuclei-lesioned and ovariectomized rats, which suggested that the pineal gland can modulate prolactin secretion by acting through a dopamine mechanism independent of hypothalamic suprachiasmatic structures.  相似文献

Normal human autopsy anterior pituitary tissue from 5 cases was embedded in LR White resin and immunolabelled using silver-enhanced 5-nm protein A gold probes. Follicle stimulating hormone (FSH), thyroid stimulating hormone (TSH), luteinizing hormone (LH), adrenocorticotrophic hormone (ACTH), growth hormone (GH) and prolactin (PRL) were immunolocalised to the level of secretory granule. A two-sided double-labelling method was used to cross-react two hormones at a time with respect to their corresponding antibodies. All possible combinations of the six pituitary hormones were tested. Plurihormonal granules were found that contained LH + FSH, LH + TSH, and FSH + TSH. Each hormone was also found in monohormonal granules. Summary Biological and statistical aspects of the application of variance to mean ratio to describe spatial distribution of animals are discussed. It is shown that the parameterb inTaylor's power laws 2=a m b shows intra-specific variation depending on the distribution of the constituent units of the population.a andb are only parameters of a very empirical way of describing the relation between variance and mean, which itself is an indicator for spatial distribution. Summary Previous studies have shown that daily afternoon injections of melatonin in anosmic male rats result in depressed accessory sex organ weights and serum prolactin levels.
