Summary The reproductive activity of virgin females of anAcanthoscelides obtectus strain originating from the high Colombian plateau was variable. Some females did not synthesize vitellogenin during imaginal life while others produced mature oocytes. In the case of virgin females without reproductive activity, the supply of pollen in the adult diet did not stimulate synthesis and incorporation of vitellogenin. On the other hand, the supply of pollen always induced a higher vitellogenin titre in the haemolymph of females which produced oocytes, and stimulated ovarian production.  相似文献

Summary Fat bodies from adult females ofLocusta migratoria continue to synthesize vitellogenin and other proteins when cultured in vitro. A strong secondary induction of vitellogenin synthesis was obtained in fat bodies cultured in the presence of methoprene, and a weaker but significant primary induction was also obtained using higher doses (>100 g) of methoprene.This work was carried out in Dr G.R. At any point in the sexual cycle of both males and females, oestradiol benzoate induces synthesis of plasmatic vitellogenin, identifiable by a specific immunserum. Pharmacological doses of purified testosterone and dihydrotestosterone also induce synthesis of this yolk precursor. Although a partial aromatisation of testosterone into oestrogen is possible, our results show that there must also be interaction between the androgens and the oestrogens receptors in the liver.  相似文献

Summary The selectivity of vitellogenin absorption by the locust oocyte was examined by comparing the uptake of vitellogenin and a haemolymph protein of similar molecular weight (MHP). Though both proteins occurred in the haemolymph at approximately the same concentration there occurred a 500-fold difference in accumulation of vitellogenin over MHP during a 24-h period. Surprisingly MHP did not accumulate in the oocyte during vitellogenesis.  相似文献

Summary The distribution of3H-leucine between the various polyopeptide components ofOrchestia gammarellus vitellogenin and lipovitellins, separated by SDS-PAGE, has been stdied after in vivo injection of this amino-acid. Summary One major antigen, present in female cuticle, female blood and eggs, is revealed by the antiserum against soluble cuticular proteins of adults locusts, and by all the antisera against vitellin or vitellogenin. It is not revealed by the specific antiserum against diglyceride-binding lipoprotein. The presence of this major antigen in the cuticle depends on the presence of vitellogenin in the blood.  相似文献

Summary The activity of the pure 10R (=natural) and 10S enantiomers of juvenile hormone III (JH III) was determined in 3 different bioassays, and the relative binding affinity of the 2 enantiomers to the haemolymph JH-binding protein of the cockroachNauphoeta cinerea was measured. In theGalleria wax test, a local morphogenetic assay, the 10R enantiomer was 5240 times more active than, the 10S enantiomer, 1Galleria unit corresponding to 0.42 pg of 10R-JH III as compared to 2.2 ng for 10S-JH III. In a systemic morphogenetic assay with the cockroachNauphoeta cinerea 380 times less 10R enantiomer was necessary in order to induce detectable juvenilisation (58 ng 10R and 22 g 10S) and in a systemic gonadotropic assay withNauphoeta cinerea 255 times less 10R was needed to induce vitellogenin synthesis in 50% of the insects (6.7 ng 10R and 1710 ng 10S). In the JH-binding protein assay 10R-JH III had an affinity for the JH-binding protein (lipophorin) which was approximately 46 times higher than that of 10S-JH III.  相似文献

Vitellogenin and lipovitellins of Orchestia gammarella, tested by electrophoresis on gels of different acrylamide concentrations, following the procedure of Hedrick and Smith (1968), displays a migration pattern identical to that of proteins of respectively congruent to 4 x 10(5) (vitellogenin), congruent to 3,5 x 10(5) (lipovitellins I and I') and congruent to 5 x 10(5) (lipovitellin II) molecular weights.  相似文献

Eleocytes are specialized coelomic cells in nereid annelids which assume a central role during germ cell development. They may contain extremely high concentrations of both adenosine monophosphate (AMP) and adenosine diphosphate (ADP) (each >10 mol/ml of cell vol.), whereas the adenosine triphosphate (ATP) content is comparatively low (0.8 mol/ml cell vol.).31P nuclear magnetic, resonance (NMR) studies of living eleocytes suggest the compartmentalization of both AMP and ADP in the large acidic vacuole characteristic for this cell type. Eleocytes are thus capable of storing high concentrations of ADP and AMP without inhibiting energy metabolism, by sequestering these compounds in a separate compartment. The high concentrations of both AMP and ADP in the eleocytes decrease in both males and females during the course of maturation. In eleocytes of male animals, the decline of the high nucleotide concentrations was accompanied by a transient increase of two intracellular nucleosides, inosine and guanosine. This suggests the degradation and further metabolism of nucleotides to the corresponding nucleosides. In culture, eleocytes release both inosine and guanosine into the medium. Both nucleosides are also present in the coelomic fluid, the common compartment for both eleocytes and germ cells. Both male and female germ cells incorporate14C-labelled inosine and guanosine in culture. For oocytes, the further incorporation of [14C]inosine into the RNA fraction could be demonstrated. The large adenylate pools in the eleocytes may be regarded as a store for purine compounds for later use by the growing germ cells to supplement nucleic acid synthesis. The supply of nucleic acid precursors seems to be another specific function of eleocytes related to gametogenesis, in addition to their known synthesis of vitellogenin.  相似文献

Summary A competitive enzyme linked immunosorbent assay was developed for the quantification of a large lipoprotein, namely the yolk protein vitellogenin, in the haemolymph of cockroaches (Nauphoeta cinerea). Summary Tamoxifen is an anticancer drug widely used in the treatment of estrogen-dependent breast cancer. In hatchling alligators it acts as a pure antiestrogen in that it completely blocks the effect of estradiol-induced oviductal hypertrophy and completely blocks the estradiol-induced hepatic vitellogenin secretion. Paradoxically, when injected into alligator eggs incubated at 33°C, a temperature which would normally result in 100% male hatchlings, tamoxifen sex reverses the embryos into apparently normal female hatchlings.  相似文献

Immunological resistance of the chick embryo is dependent upon IgG present in the yolk of the layed egg. Here we show that complement factor 3 (C3), a key component of the humoral complement system, is a yolk component of chicken eggs. C3 is transported into oocytes by LR8-mediated endocytosis. LR8 also binds and transports other major yolk components such as vitellogenin, very-low-density lipoprotein, and α2-macroglobulin. Expression studies of LR8 during chicken development and oocyte maturation, in combination with studies on the uptake of individual yolk components, suggest the following model for oocyte maturation in the chicken: all oocytes present in the ovary contain high levels of LR8 mRNA and protein long before the onset of oocyte maturation. Selected oocytes gain access to yolk precursors, and LR8 binds, internalizes, and deposits the major yolk components in the ratio of their relative abundance in the accessible pool.Received 9 May 2005; received after revision 6 June 2005; accepted 13 June 2005  相似文献

The relationship between incorporation of intravenously injected 14C lysine and specific radio-activity of precursor was used to estimate protein synthesis in muscle of growing lambs. The rate of protein synthesis per unit of muscle weight in Supraspinatus and Extensor digitorum longus decreased strongly from one week of age to puberty (10 weeks); afterwards it decreased in supraspinatus and increased slightly in Extensor digitorum longus. The rate of protein synthesis increase in muscle protein weight was constant during the whole experiment (1 week-16 weeks). In preruminant Lambs )1 week-5 weeks) the rate of protein synthesis per unit of muscle weight decreased; however, due to the increase in muscle weight, the rate of protein synthesis in whole muscle remained relatively constant. In order Lambs the rate of protein synthesis in whole muscle decreased. The turnover time of protein increased with age. These results give some explanation on muscular development of Lambs.  相似文献

Summary DNA and cholesterol synthesis were investigated in the kidneys of fasted-refed rats. Refeeding resulted in an increase in kidney DNA synthesis, as measured by3H-thymidine incorporation, starting at 72 h. Either mitomycin C or cytosine-arabinoside caused cessation of antigen-containing cell number increase. Stimulation of cellular DNA synthesis by growth medium change increased the level of antigen-containing cells. Replicative DNA synthesis in permeable mouse sarcoma cells was more sensitive to ethidium bromide (EtBr) than bleomycin-induced unscheduled DNA synthesis (UDS). A similar difference in sensitivity to EtBr was observed between DNA polymerases alpha and beta. The difference in sensitivity to EtBr of replicative DNA synthesis and UDS in the present system seems to reflect mainly the sensitivity difference between DNA polymerases alpha and beta.  相似文献

In this study, earlier observations concerning the independence of both natural (NCMC) and lectin-dependent cell-mediated cytotoxicity (LDCC) from DNA synthesis have been confirmed. In addition, blocking of RNA synthesis by actinomycin D and of protein synthesis, reversibly by puromycin (PM) and irreversibly by emetine (EM) had different effects on NCMC and LDCC against 3H-thymidine-prelabeled HEp-2 target cells. Similarly to the Con A-induced proliferation of lymphocytes, LDCC activity was also inhibited by blocking of RNA and protein synthesis. NCMC to HEp-2 target cells was not affected by blocking of RNA synthesis, while both PM and EM strongly enhanced NCMC activity.  相似文献
