山茱萸多糖提取过程研究

摘    要:通过单因素实验及正交实验对提取液中多糖含量的各种影响因素进行了研究,确定了山茱萸多糖最佳提取工艺条件:提取温度 80℃,提取时间 2h,料液比1∶16,原材料粒度 300~450μm,提取次数4次,脱蛋白次数 6次。在此条件下提取并用DEAE-cellulose柱层析分离得到白色多糖PFCAⅢ,经IR检测为含有α糖苷键的多糖。

Abstract:In this paper, several factors of affecting the extraction process were studied through orthogonal experiments of L9(3)4. The optimal technological conditions of water extraction from Frutus Corni were decided: the temperature being 80℃, the extraction time being 2h, the ratio of solid material to water being 1∶16, the granularity of Frutus Corni being 300~450μm, the times of extraction at the same ratio of solid material to water being 4, and the times of clearing protein being 6. The purified product which was prepared with chromatographic fractionating of DEAE-cellulose was proved to be polysaccharide by IR.
