三维培养基质体系在细胞迁移研究中的应用

摘    要:细胞迁移的研究中可用到的培养基质远不能满足需要.利用鼠尾胶和基底膜胶质(Matrigel)混合形成三维Collagen-Matrigel混合胶,该混合胶在培养液∶鼠尾胶∶基底膜胶质为1∶5∶4配比下,0.5 h内凝固满足组织培养要求.同时该混合胶经固定后在光学显微镜下结构维持良好,无颗粒.弥散性因子表达细胞团块和上菱唇组织块共培养于该三维Collagen-Matrigel混合胶培养基质中,结果显示细胞迁移良好,SDF-1因子和Slit因子分别对小脑上菱唇细胞具有化学吸引作用和化学排斥作用.

Abstract:The matrix used in cell culture cannot match the requirement during the cell migration research in vitro at present.The three-dimentional collagen-matrigel mixture gel was studied which was made up of culture medium: collagen: matrigel by different ratios.The mixture gel got solidified within half an hour at room temperature,and did not show visible particles under microscope observation after fixation.The results from co-culture of cell aggregate and upper rhombic lip (URL) explant indicated that SDF-1 has the chemoattraction to URL cells,while slit has the chemorepulsion to them.
