白色念珠菌与细菌混合感染小鼠阴道炎动物模型的构建及感染菌的鉴定

摘要: 摘要: 目的 在制作单一白色念珠菌性阴道炎动物模型的过程中,出现了两例严重感染的小鼠,因此,对其感染机 制进行了深入研究。方法 从严重感染小鼠的阴道内分离得到真菌与细菌,经纯化 3 代后得到 15 个真菌样本和 19 个细菌样本,对于真菌样本进行念珠菌的显色培养基鉴定,对于细菌进行了 16SrDNA 提取、PCR 验证和 PCR- DGGE 分析,对不同种细菌的 DNA 样本进行序列测定。结果 分离出 1 种真菌和 4 种致病性细菌,证实了小鼠阴 道重度感染是由 1 种真菌和 4 种细菌共同作用而引起。结论 我们所发现的小鼠阴道重度感染模型,是由白色念 珠菌( Monilia albican) 与 4 种细菌,即变形菌( Proteus vulgaris) 、嗜麦芽寡养单胞菌( Stenotrophomonas maltophilia) 、产 碱杆菌( Alcaligenes Castellani&Chalmers) 和芽孢杆菌( Bacillus) 的混合感染所致。

Abstract: Abstract: Objective In the process of making animal models with single Monilia albicans vaginitis,there were two cases of serious infection in mice,so the infection mechanism were studied deeply. Method Isolating fungi and bacteria from mice vagina with severe infection,the 15 fungal samples and 19 samples of bacteria were got after purifying for 3 generations. For fungal samples,we conducted chromogenic medium identification of Monilia albicans. For samples of bacteria,we conducted 16SrDNA extraction,PCR validation and PCR-DGGE analysis, besides we conducted sequence determination of different bacteria DNA samples. Result We isolated 1 kind of fungi and 4 kinds of pathogenic bacteria,it confirmed that the mouse vaginal severe infection is caused by the interaction of 1 kind of fungi and 4 kinds of bacteria. Conclusion The mouse vaginal severe infection model we found,is caused by the mixed infection of 1 kind of fungi: Monilia albican and 4 kinds of bacteria: Proteus vulgaris,Stenotrophomonas maltophili,Alcaligenes Castellani&Chalmers and Bacillus.
